摘要
目的:探讨芍药苷(PAE)调节丝氨酸/苏氨酸激酶(AKT)/鼠双微基因2(MDM2)/p53信号通路对弥漫大B细胞淋巴瘤(DLBCL)细胞增殖、凋亡和细胞周期的影响。方法:以OCI-LY3细胞为研究对象,分别设置PAE低浓度组(15μmol/L PAE)、PAE中浓度组(30μmol/L PAE)、PAE高浓度组(60μmol/L PAE)、PAE高浓度+SC79(AKT激活剂)组(60μmol/L PAE+8μg/ml SC79),同时以未经处理的细胞为对照组。48 h后,分析细胞增殖、克隆形成能力及细胞周期和凋亡变化,检测AKT/MDM2/p53信号通路相关蛋白的表达水平。结果:与对照组比较,PAE低浓度组、PAE中浓度组、PAE高浓度组细胞凋亡率、p53表达、G_(0)/G_(1)期增加,细胞克隆形成数和细胞增殖率,p-AKT/AKT和p-MDM2/MDM2表达水平,以及G_(2)/M、S期降低(均P<0.05)。与PAE高浓度组比较,PAE高浓度+SC79组细胞凋亡率、p53表达、G_(0)/G_(1)期降低,细胞克隆形成数和细胞增殖率,p-AKT/AKT和p-MDM2/MDM2表达水平,以及G_(2)/M、S期增加(均P<0.05)。结论:PAE通过抑制AKT/MDM2上调p53表达,抑制DLBCL细胞增殖、细胞周期进展,诱导其凋亡。
Objective:To investigate the effects of paeoniflorin on proliferation,apoptosis and cell cycle of diffuse large B-cell lymphoma cells by regulating serine/threonine kinase(AKT)/murine double minute 2(MDM2)/p53 signaling pathway.Methods:OCI-LY3 cells were regarded as the research object,and PAE low concentration group(15μmol/L PAE),PAE medium concentration group(30μmol/L PAE),PAE high concentration group(60μmol/L PAE),and PAE high concentration+SC79(AKT activator)group(60μmol/L PAE+8μg/ml SC79)were set,respectively.At the same time,untreated cells were regarded as the control group.After 48 hours,cell proliferation,colony formation ability,cell cycle and apoptosis were analyzed,and the expression levels of AKT/MDM2/p53 signaling pathway related proteins were detected.Results:Compared with the control group,the apoptosis rate,p53 expression,G_(0)/G_(1) phase of PAE low concentration group,PAE medium concentration group and PAE high concentration group were obviously increased,the number of cell clones formed,proliferation rate,the expressions of p-AKT/AKT and p-MDM2/MDM2,the G_(2)/M and S phases were obviously reduced(all P<0.05).Compared with the PAE high concentration group,the apoptosis rate,p53 expression,G_(0)/G_(1) phase of the PAE high concentration+SC79 group were obviously decreased,the number of cell clones formed,proliferation rate,the expressions of p-AKT/AKT and p-MDM2/MDM2,the G_(2)/M and S phases were obviously increased(all P<0.05).Conclusion:PAE can inhibit DLBCL cell proliferation,cell cycle progression,and induce apoptosis by inhibiting AKT/MDM2 expression and up-regulating p53 expression.
