摘要
目的:探讨芪黄疽愈方调控巨噬细胞相关因子影响ApoE^(-/-)小鼠动脉粥样硬化形成的作用机制。方法:68只ApoE^(-/-)小鼠随机分为模型组(24只)、中药组(22只)、对照组(22只),另设C57BL/6小鼠25只为空白组。高脂饲料喂养构造动脉粥样硬化ApoE^(-/-)小鼠模型,中药组按0.2 mL/20 g体质量予芪黄疽愈方颗粒灌胃,对照组按0.2 mL/20 g体质量予阿托伐他汀钙片悬混液灌胃,空白组、模型组给予等量0.9%氯化钠溶液灌胃,连续给药12周。观察各组小鼠一般状态、体质量;主动脉HE染色、Masson染色、油红O染色,观察血管病理形态、斑块面积;血清ELISA检测巨噬细胞相关因子白细胞介素(IL)-1β、IL-4、IL-13、干扰素-γ(IFN-γ)、脂多糖(LPS)、转化生长因子-β(TGF-β)、IL-6、IL-10、IL-12表达情况;免疫荧光观察巨噬细胞表面标志物iNOS、CD206表达情况。结果:模型组、中药组、对照组小鼠精神萎靡,毛色暗淡,前期增长迅速,后逐渐减缓,其中模型组体质量显著高于其他各组(P<0.01)。HE染色、Masson染色、油红O染色病理图片显示:模型组较空白组斑块面积显著增大,胶原纤维含量明显减少,主动脉红染脂质显著增加(P<0.01),中药组、对照组较模型组斑块面积降低,胶原纤维含量明显增多,主动脉红染脂质显著减少(P<0.01)。ELISA检测结果示:模型组小鼠血清IL-1β、IFN-γ、LPS、IL-6、IL-12显著高于空白组(P<0.05),IL-4、IL-13、TGF-β、IL-10显著低于空白组(P<0.05);与模型组比较,中药组小鼠血清IL-1β、IFN-γ、LPS、IL-6、IL-12显著降低(P<0.05),IL-4、IL-13、TGF-β、IL-10的表达水平显著提高(P<0.05)。免疫荧光染色示:M1型巨噬细胞标志物iNOS在模型组阳性表达较空白组高(P<0.01),中药组表达较模型组低(P<0.05),M2型巨噬细胞标志物CD206在模型组阳性表达较中药组低(P<0.01)。结论:芪黄疽愈方可减少M1型巨噬细胞,抑制M1型巨噬细胞相关因子IL-1β、IFN-γ�
Objective:To explore the mechanism of Qihuang Juyu Formula affecting atherosclerosis by regulating ApoE^(-/-)mice macrophage related cytokines.Methods:Sixty-eight ApoE^(-/-)mice were randomly divided into a model group(n=24),a traditional Chinese medicine group(n=22),and a control group(n=22),with 25 C57BL/6 mice as the blank group.The atherosclerotic ApoE^(-/-)mouse model was constructed by feeding high-fat diet.In the Chinese medicine group,Qihuang Juyu Formula Granules were administered to mice by gavage with 0.2 mL/20 g body weight.In the control group,atorvastatin calcium tablet suspension was administered to mice by gavage with 0.2 mL/20 g body weight.The blank group and model group were given an equal amount of 0.9%sodium chloride solution by gavage.All were continuously administered for 12 weeks.Observe the general state and body mass of each group of mice.Aortic HE staining,Masson staining,and oil red O staining were used to observe the pathological morphology and plaque area of the blood vessels.Serum ELISA detection of macrophage related factors expression,including interleukin(IL)-1β,IL-4,IL-13,interferon-γ(IFN-γ),Lipopolysaccharides(LPS),transforming growth factors-β(TGF-β),IL-6,IL-10,IL-12.Immunofluorescence was used to observe the expression of macrophage surface markers iNOS and CD206.Results:The mice in the model group,traditional Chinese medicine group,and control group showed signs of mental fatigue,dull hair color,rapid growth in the early stage,and gradually slowing down later.The quality of the model group was significantly higher than that of other groups(P<0.01).The pathological images of HE staining,Masson staining,and Oil Red O staining showed that the model group had a significant increase in plaque area,a significant decrease in collagen fiber content,and a significant increase in aortic red stained lipids compared to the blank group(P<0.01).The traditional Chinese medicine group and control group had a significant decrease in plaque area,an increase in collagen fiber content,and
