间充质干细胞膜包载5-氨基酮戊酸介导的光热作用抑制皮脂分泌

摘要 目的探究间充质干细胞膜包载5-氨基酮戊酸(ALA@STCM)介导的光热作用对金黄地鼠皮脂腺斑皮脂分泌的影响。方法培养脐带来源的间充质干细胞(MSCs),提取MSC膜,制备ALA@STCM。首先,通过CCK-8实验对比5-氨基酮戊酸(ALA)和ALA@STCM对皮脂腺细胞活力的影响。其次,建立金黄地鼠痤疮动物模型,通过组织学分析对比ALA和ALA@STCM对皮脂分泌的影响。结果CCK-8结果表明,相较于ALA,使用MSC膜包载ALA未明显增加细胞毒性,证明了MSC膜作为载药工具的安全性。组织学分析结果表明,给予光照后,ALA和ALA@STCM均能不同程度地缩小皮脂腺斑面积和抑制皮脂分泌,但是我们发现ALA@STCM的缩小皮脂腺斑的作用和抑制皮脂分泌作用明显优于ALA。结论ALA@STCM介导的光热作用可缩小金黄地鼠的皮脂腺斑、抑制皮脂分泌,并且显著优于ALA。 Objective This study aimed to explore the impact of photothermal therapy mediated by mesenchymal stem cell membrane-coated 5-aminolevulinic acid(ALA@STCM) on sebum secretion in sebaceous gland hyperplasia.Methods Mesenchymal stem cells(MSCs) derived from umbilical cords were cultured, and MSC membranes were extracted to prepare ALA@STCM.Firstly, the effect of 5-aminolevulinic acid(ALA) and ALA@STCM on the viability of sebaceous gland cells was compared using a CCK-8 assay.Secondly, a golden hamster acne animal model was established, and histological analysis was conducted to compare the effects of ALA and ALA@STCM on sebum secretion.Results The CCK-8 assay results indicated that, compared to ALA,the use of MSC membrane-coated ALA did not significantly increase cytotoxicity, confirming the safety of MSC membrane as a drug delivery tool.Histological analysis results revealed that, following light exposure, both ALA and ALA@STCM could reduce sebaceous gland hyperplasia and inhibit sebum secretion to varying degrees.However, it was observed that ALA@STCM had a significantly greater effect in reducing sebaceous gland hyperplasia and inhibiting sebum secretion compared to ALA alone.Conclusion Photothermal effects mediated by ALA@STCM can effectively reduce sebaceous gland hyperplasia in golden hamsters, inhibit sebum secretion, and exhibit a significant advantage over ALA in achieving these effects.
