白玉菇多糖理化性质及免疫活性研究

摘要 文章采用常温水提和热水浸提法从白玉菇子实体中提取常温水提多糖(WHN)和沸水浸提多糖(WHB),通过DEAE-Cellulose离子交换层析柱进行分离纯化获得6种分级组分WHN_(1)、WHN_(2)、WHN_(3)、WHB_(1)、WHB_(2)、WHB_(3)。理化性质结果表明:WHN和WHB的总糖质量分数为67.72%和80.54%,分级组分总糖质量分数为93.48%~98.77%,含有少量蛋白质和糖醛酸;多糖组分的单糖摩尔分数各不相同,WHN、WHN_(1)、WHN_(2)、WHN_(3)是以半乳糖和葡萄糖为主的杂多糖,WHB、WHB_(1)、WHB_(2)、WHB_(3)组分中葡萄糖占比均最大,WHB_(3)为葡聚糖。采用药理学模型考察多糖对鼠巨噬细胞增殖活性、吞噬活性以及NO释放量的影响,结果表明,多糖WHN_(1)和WHB_(3)在体外细胞实验中表现出较好的免疫调节活性。研究结果为白玉菇多糖在药物和功能性食品中的应用提供理论基础。 In this study,the polysaccharides WHN and WHB were isolated from the fruiting bodies of white Hypsizygus marmoreus by water extraction at normal and boiling temperatures.After purification by DEAE-Cellulose ion exchange chromatography,six fractions WHN_(1),WHN_(2),WHN_(3),WHB_(1),WHB_(2) and WHB_(3) were obtained.The results of physicochemical properties showed that the total sugar content of WHN and WHB was 67.72%and 80.54%,while the total sugar content of fractions ranged from 93.48%to 98.77%.The content of protein and uronic acid was little.The mole fraction of monosaccharide in eight components was different.WHN,WHN_(1),WHN_(2) and WHN_(3) were heteropolysaccharides dominated by galactose and glucose,and the highest proportion of glucose was found in WHB,WHB_(1),WHB_(2) and WHB_(3),among which WHB_(3) was glucan.Pharmacological models were used to investigate the effects of polysaccharides on the proliferation activity,phagocytosis activity and NO release of macrophages in mice.It was revealed that polysaccharides WHN_(1) and WHB_(3) showed better immunomodulatory activities in in vitro cell experiments.The results provide a theoretical basis for the application of white Hypsizygus marmoreus polysaccharides in medicine and functional food.
