六星瓢虫线粒体全基因组测序分析及系统发育研究

摘要 以巧瓢虫属的捕食性天敌六星瓢虫为研究对象,测定了六星瓢虫的线粒体全基因组,通过比较分析明确了六星瓢虫的线粒体基因组特征,并通过构建系统发育树论证了瓢虫科的系统发育关系。结果表明:(1)六星瓢虫线粒体基因组全长17885 bp(GenBank登录号:OR804096),拥有37个基因,包括13个PCGs、2个rRNA基因、22个tRNA基因以及1个控制区,基因排列序列与其他已发布的瓢虫科昆虫的线粒体基因组一致,无重排现象;(2)六星瓢虫线粒体基因组整体AT含量为79.1%,蛋白质编码基因使用频率最高的前3位密码子分别是:UUA、AUU和UUU;(3)基于13 PCGs+2 rRNAs基因序列数据集的最大似然树结果表明,各瓢虫亚科的单系性均得到了支持,瓢虫科内部的系统发育关系为(Sticholotidinae+((Epilachninae+Scymninae)+Coccinellinae));(4)新测序的六星瓢虫与六斑异瓢虫的关系最近,在瓢虫族内部,支持((Anatis+(Calvia+Propylea))+((Hippodamia+Harmonia)+(Cycloneda+(Coccinella+(Oenopia+Aiolocaria)))))的近缘关系。综上,研究丰富了瓢虫科线粒体基因组数据,为解决瓢虫科的系统发育关系提供了参考。 The predatory predator ladybird Oenopia formosana within the genus Oenopia was taken as the research object,and the mitochondrial genome of O.formosana was determined.The mitochondrial genome characteristics of O.formosana were defined by comparative analysis,and the phylogenetic relationship of Coccinellidae was demonstrated by constructing phylogenetic trees.The results showed that:(1)The total length of mitochondrial genome of O.formosana was 17885 bp(GenBank entry number:OR804096),containing 37 genes,including 13 PCGs,2 rRNA genes,22 tRNA genes and a control region,was consistent with other published mitochondrial genomes of Coccinellidae,without rearrangement.(2)The AT content of the mitochondrial genome was 79.1%,and the top three codons used most frequently by protein-coding genes were UUA,AUU and UUU.(3)The maximum likelihood tree results based on the 13PCGs+2rRNAs gene sequence data set showed that the monophylety of each subfamily within Coccinellidae was supported.The phylogenetic relationship within the family is(Sticholotidinae+((Epilachninae+Scymninae)+Coccinellinae)).(4)The newly sequenced O.formosana was most closely related to the ladybird Aiolocaria hexaspilota.Within Coccinellini,The phylogenetic relationship is((Anatis+(Calvia+Propylea))+((Hippodamia+Harmonia)+(Cycloneda+(Coccinella+(Oenopia+Aiolocaria))))).This study enriches the mitochondrial genome data of Coccinellidae and provides a reference for resolving the phylogenetic relationships of Coccinellidae.
