地榆七柏汤含药血清对H_(2)O_(2)诱导的Caco-2细胞Th1/Th2平衡的影响

摘要 目的:分析地榆七柏汤含药血清对H2O2诱导的Caco-2细胞Th1/Th2平衡的影响,为地榆七柏汤的临床开发和机制研究提供实验依据。方法:选取60只健康雄性SD大鼠,将其随机分为空白组、模型组、柳氮磺胺吡啶(SASP)组、地榆七柏汤组(分低、中、高剂量3组),每组10只。构建三硝基苯磺酸(TNBS)诱导的溃疡性结肠炎(UC)动物模型,空白组和模型组均给予生理盐水灌肠,SASP组、地榆七柏汤组(分低、中、高剂量3组)分别给予含SASP、生药(剂量为0.5g/ml、1.0g/ml、3.0g/ml)的汤剂灌肠,制备相关血清,以H_(2)O_(2)为刺激因子,以Caco-2细胞为载体,建立Caco-2炎症细胞模型,检测对比各组白介素(IL)-4、干扰素(INF)-γ及INF-γ/IL-4水平。结果:大鼠DAI评分:模型组>SASP组>低剂量地榆七柏汤组>中剂量地榆七柏汤组>高剂量地榆七柏汤组>空白组(均P<0.05)。大鼠血清INF-γ、INF-γ/IL-4水平:模型组>SASP组>低剂量地榆七柏汤组>中剂量地榆七柏汤组>高剂量地榆七柏汤组>空白组(均P<0.05);血清IL-4水平:模型组<SASP组<低剂量地榆七柏汤组<中剂量地榆七柏汤组<高剂量地榆七柏汤组<空白组(均P<0.05)。Caco-2细胞INF-γ、INF-γ/IL-4水平:细胞模型组>SASP组>低剂量地榆七柏汤组>中剂量地榆七柏汤组>高剂量地榆七柏汤组>空白对照组(均P<0.05);Caco-2细胞IL-4水平:细胞模型组<SASP组<低剂量地榆七柏汤组<中剂量地榆七柏汤组<高剂量地榆七柏汤组<空白对照组(均P<0.05)。结论:地榆七柏汤含药血清能够下调H_(2)O_(2)诱导的Caco-2细胞中Th1细胞因子(INF-γ)分泌和上调Th2细胞因子(IL-4)水平,促使Th1/Th2平衡恢复,起到抗炎效果,且高剂量地榆七柏汤含药血清效果更佳。 Objective:To analyze the effects of Diyu Qibai decoction-containing serum on Th1/Th2 balance of H_(2)O_(2)-induced Caco-2 cells,and to provide experimental evidence for the clinical development and mechanism research of Diyu Qibai decoction.Methods:Sixty healthy male SD rats were selected and randomly divided into blank group,model group,sulfasalazine(SASP)group and Diyu Qibai decoction groups(low-,middle-and high-dose groups),with 10 rats in each group.The animal models of trinitrobenzene sulfonic acid(TNBS)-induced ulcerative colitis(UC)were constructed.Blank group and model group were given normal saline enema,and SASP group and Diyu Qibai decoction groups(low-,middle-and high-dose groups)were given decoction enema containing SASP and crude drug(doses of 0.5g/ml,1.0g/ml,3.0g/ml)respectively.The related serum was prepared,and H_(2)O_(2)was set as stimulating factor and Caco-2 cells as carriers to establish the Caco-2 inflammatory cell model,and the levels of interleukin(IL)-4,interferon(INF)-γand INF-γ/IL-4 in each group were detected and compared.Results:DAI score of rats showed that model group>SASP group>low-dose Diyu Qibai decoction group>middle-dose Diyu Qibai decoction group>high-dose Diyu Qibai decoction group>blank group(all P<0.05).The levels of serum INF-γand INF-γ/IL-4 of rats showed that model group>SASP group>low-dose Diyu Qibai decoction group>middle-dose Diyu Qibai decoction group>high-dose Diyu Qibai decoction group>blank group(all P<0.05).Serum IL-4 level showed that model group<SASP group<low-dose Diyu Qibai decoction group<middle-dose Diyu Qibai decoction group<high-dose Diyu Qibai decoction group<blank group(all P<0.05).The levels of INF-γand INF-γ/IL-4 in Caco-2 cells showed that model group>SASP group>lowdose Diyu Qibai decoction group>middle-dose Diyu Qibai decoction group>high-dose Diyu Qibai decoction group>blank control group(all P<0.05).The IL-4 level inCaco-2 cells showed that cell model group<SASP group<low-dose Diyu Qibai decoction group<middle-dose Diyu Qibai decoction group
