摘要
目的构建超高效液相色谱串联三重四级杆质谱法(UPLC⁃MS/MS)同时测定华重楼抽芽前期根茎9种植物内源激素的含量,为其高效栽培技术提供依据。方法色谱方法:采用Waters CORTECS C_(18)色谱柱(2.1 mm×100 mm,1.6μm),流动相乙腈(A)⁃0.1%甲酸水(B);流速为0.3 mL·min^(-1),梯度洗脱,柱温40℃,进样量2μL。质谱方法:采用ESI电喷雾离子源,以正负离子切换多反应监测(MRM)模式进行含量测定。结果9种内源激素在考察的浓度范围内呈良好的线性关系(r>0.999);加样回收率在70.26%~115.85%,RSD为0.45%~6.2%;脱落酸(ABA)、赤霉素4(GA4)、茉莉酸(JA)和异戊烯基腺嘌呤(IP)含量较低,而赤霉素7(GA7)、顺式玉米素(CZ)、反式玉米素核苷(TZR)、吲哚乙酸(IAA)和水杨酸(SA)含量较高。结论该方法操作简便、快速、准确,可用于华重楼根茎内源激素的含量测定;GA7、CZ、TZR、IAA、SA等植物内源激素共同调控华重楼根茎生长发育,为促进其根茎休眠解除与繁殖的栽培技术提供理论依据。
OBJECTIVE An ultra⁃high performance liquid chromatography tandem triple quadrupole mass spec⁃trometry(UPLC⁃MS/MS)method was developed to simultaneously determine the contents of 9 endogenous hormones in the early stage of sprouting of Paris polyphylla rhizome,providing a basis for its efficient cultivation tech⁃niques.METHODS The chromatography was performed on a Waters CORTECS C_(18)column(2.1 mm×100 mm,1.6μm)with acetonitrile(A)and 0.1%formic acid water(B)as mobile phases.The flow rate was 0.3 mL·min^(-1)in gradient elution,the column temperature was 40℃and the injection volume was 2μL.Mass spectrometry method:ESI electrospray ion source was used for contents determination under positive and negative ion switching multiple reaction monitoring(MRM)mode.RESULTS The contents of 9 endogenous hormones showed a good lin⁃ear relationship(r>0.999)within the investigated concentration range,the average recovery ranged from 70.26%⁃115.85%with the RSD ranged from 0.45%⁃6.2%.The contents of abscisic acid(ABA),gibberellin 4(GA4),jas⁃monic acid(JA)and isopentenyladenine(IP)were low,while the contents of gibberellin 7(GA7),Cis⁃zeatin(CZ),trans zeatin nucleoside(TZR),indole acetic acid(IAA)and salicylic acid(SA)were high.CONCLUSION This UPLC⁃MS/MS method is simple,speedy,and accurate,which can be used to determine the contents of endog⁃enous hormones in the rhizomes of Paris polyphylla.Endogenous hormones such as GA7,CZ,TZR,IAA,SA,etc.jointly regulate the growth and development of the rhizomes of Paris polyphylla,providing a theoretical basis for promoting the release of dormancy and the reproductive cultivation techniques of its rhizomes.
