摘要
为了筛选桔梗种子生活力测定的适宜条件,建立其生活力测定的快速方法,以桔梗种子为试验材料,采用2,3,5-氯化三苯基四氮唑(TTC)染色法,先通过单因素试验确定生活力测定的影响因素及其范围,再通过Box-Benhnken响应面法试验,确定TTC浓度、染色温度、染色时间对桔梗种子染色影响的最佳条件。结果表明,桔梗种子染色的最佳条件为:TTC浓度为0.90%,染色温度为45℃,染色时间为16 h,其影响程度为:染色温度>染色时间>TTC浓度。在此优化条件下桔梗种子生活力理论值为76.05%,实际测定值和理论预测值的误差为0.7%(<5%),说明响应面法优化桔梗种子TTC法测定条件是可行的,利用TTC法了解桔梗种子潜在发芽能力,为桔梗种子生产及其特性的研究提供依据。
To screening the appropriate conditions and to establish a rapid method for the determination of seed viability of Platycodon grandiflorus,and seeds were stained with 2,3,5-triphenyl tetrazolium chloride(TTC)using Platycodon grandiflorus seeds as materials,and the influencing factors and value range were first determined by single-factor test,and then the effects of TTC concentration,staining temperature and staining time on the staining of the seed were determined by Box-Benhnken response surface test.The optimal conditions for the staining was TTC concentration of 0.90%,staining temperature of 45℃,and staining time of 16 hours.The degree of influence was:staining temperature>staining time>TTC concentration.The theoretical value of the seed under the optimized conditions was 76.05%,and the error between the actual measured value and the theoretical predicted value was 0.7%(<5%),indicating that the response surface method was feasible to optimize the conditions for the determination of the seed by the TTC method,which could be used to understand the potential germination ability and provide a basis for the study of production and characteristics for Platycodon grandiflorus seeds.
