摘要
为了解芍药种子转录组中简单重复序列SSR的分布特点,充分开发SSR分子标记,本研究对芍药种子进行转录组测序、SSR位点分析与引物设计。结果表明,芍药种子转录组共获得63992条Unigene,总长64666285 bp,搜索得到11637个SSR位点,发生频率14.94%。在所有SSR位点中有6种重复类型,以单核苷酸(51.64%)和二核苷酸(32.48%)重复类型为主,其中A/T类型和AG/CT类型SSR位点数占SSR总数的71.03%。SSR重复次数范围为5~11次,其中以5次和10次为主。SSR序列长度主要集中在10~20 bp,占总数的84.04%。针对SSR位点,共有10925个SSR位点符合要求,成功率为93.88%,共设计出32775对引物。芍药种子转录组SSR位点丰富,该研究可为种质资源的挖掘与保护、优良品种选育和数量性状基因座定位提供参考。
In order to understand the distribution characteristics of simple repeat sequence SSR in the transcriptome of Paeonia lactiflora Pall.seeds and fully develop SSR molecular markers,in this study,transcriptome sequencing,SSR site analysis and primer design were carried out for the seeds of Paeonia lactiflora Pall.The results showed that a total of 63992 Unigenes with a total length of 64666285 bp were obtained from the transcription group of Paeonia lactiflora Pall.seeds,and 11637 SSR sites were searched,with a frequency of 14.94%.Among all SSR sites,there were 6 types of repeats,mainly single nucleotide(51.64%)and dinucleotide(32.48%).The number of SSR sites of A/T type and AG/CT type accounted for 71.03%of the total number of SSRs.The number of SSR repeats was mainly distributed in 5-11 times,of which 5 and 10 times were the main ones.The length of SSR sequence is mainly 10-20 bp,accounting for 84.04%of the total.For SSR loci,a total of 10925 SSR loci met the requirements,with a success rate of 93.88%.A total of 32775 pairs of primers were designed.There were abundant SSR loci in the transcriptome of Paeonia lactiflora Pall.seeds.This study could provide reference for the excavation and protection of germplasm resources,the selection of excellent varieties and the location of quantitative trait loci.
