摘要
香豆酸3-羟化酶广泛存在于植物中,属于细胞色素P450家族的CYP98A亚家族。该酶能在植物苯丙素代谢中催化苯环C3位置的羟基化反应,是绿原酸生物合成的关键酶之一。本文以金银花香豆酸3-羟化酶基因LjC3H2为研究对象,采用生物信息学方法进行分析。结果表明,LjC3H2无信号肽,无跨膜结构域,偏亲水,定位于内质网上;二级结构以α螺旋为主,无规则卷曲较多。LjC3H2与其他植物同源蛋白氨基酸序列比对和系统发育分析表明,LjC3H2与蓟、桔梗的C3H亲缘关系较近,与同属于金银花的LjC3H亲缘关系较远;该蛋白的三级结构与细胞色素P450三级结构相似。
Coumaric acid 3-hydroxylase is widely present in various plants and belongs to the CYP45098A subfamily of the cytochrome P450 family.This enzyme can catalyze the hydroxylation reaction at the C3 position of the benzene ring in plant phenylpropanoid metabolism and is one of the key enzymes in the biosynthesis of chlorogenic acid.This article takes Lonicera japonica coumaric acid 3-hydroxylase gene LjC3H2 as the object,and uses bioinformatics methods to analyze the enzyme.the results showed that LjC3H2 has no signal peptide,no transmembrane domain,was hydrophilic,and is located on the endoplasmic reticulum.Secondary structure withαMainly spiral,with more irregular curls.Comparing this enzyme with other plant homologous enzyme proteins and conducting phylogenetic analysis,it was found that LjC3H2 was closely related to the C3H of thistle and platycodon grandiflorum,it was far-related to LjC3H,which also belongs to Lonicera japonica.The tertiary structure of this enzyme is similar to that of cytochrome P450.
