药用菊花种质遗传多样性和群体结构分析 被引量:1

摘要 对不同地理来源的64份药用菊花种质进行遗传多样性和群体结构特征分析。结果表明,从80条SCoT引物中共筛选出20条稳定性好、多态性高的SCoT引物,20条引物在64份种质间共检测出129个等位基因变异,平均每个引物可检测6.45个,变化范围为3~9个,其中SCoT29和SCoT8标记等位基因数量最多;PIC值变化范围为0.571~0.866,其中有18条SCoT标记的PIC值大于0.700。有效等位基因数范围为1.418~1.774,均值为1.583;Nei's多样性指数为0.344~0.427,平均值为0.278,Shannon's多样性指数为0.641~0.892,平均值为0.440。群体遗传多样性显示,江苏群体遗传多样性最高,河南次之,山西最低。群体间遗传距离分析表明,河北与河南、江苏和安徽、山西和江西亲缘关系较近。群体遗传结构分析表明,当K=4时,ΔK最高,群体间存在基因交流现象,基因交流程度较高。聚类分析结果表明,64份材料在0.6处被划分为四大类群,类群间的菊花种质来源地并没有明显地分开。该研究明确了64份药用菊花种质的亲缘关系,为指导亲本选配和高效育种提供重要依据。 Twenty highly polymorphic and stably amplifying markers were selected from 80 SCoT primers,which was used for PCR amplification of 64 Chrysanthemum morifolium Ramat.samples to analyse genetic diversity and population structure characteristics.The results showed that 129 variation alleles were detected from 64 germplasm resources by 20 primers,with an average of 6.45 alleles for each primers,and a variation range of 3-9,of which the primer SCoT29 and SCoT8 marked the highest number of alleles.The PIC value of each locus varied from 0.571 to 0.866,19 SCoT primers of PIC value greater than 0.700.The number of effective alleles ranged from 1.418 to 1.774,with an average of 1.583.Nei's diversity index ranged from 0.344-0.427,with an average value of 0.278,and Shannon's diversity index ranged from 0.641 to 0.892,with an average value of 0.440.The genetic diversity of Chrysanthemum morifolium Ramat.was the highest in Jiangsu followed by He'nan,and the lowest was found in Shanxi.Genetic distances among population showed that Hebei and He'nan,Jiangsu and Anhui,Shanxi and Jiangxi in relationship were near with each other.Population genetic structure showed thatΔK was the largest when K=4,there was gene exchanges between populations and high degree gene exchange was found in different populations.The results of cluster analysis showed that 64 germplasms could be classified into 4 groups at 0.6 and there were no signficant separating sources,no correlativity with the germplasm sources.There were no sharp line separating sources from groups.The study specifically genetic relationship in 64 Chrysanthemum morifolium Ramat.germplasms,it supplied an important basis for direct selection of parents and efficient breeding.
