摘要
紫花苜蓿WL712为美国引入品种,在我国南方地区已有一定范围的种植,具有高产量和高品质的优点,但该品种对部分非生物胁迫具有较差的耐受性。为了探究该品种对南方地区不同胁迫的响应,本研究以生长21 d的WL712幼苗为试验材料,对其模拟了5种南方常见的胁迫,分别为高温、低温、干旱、渍水以及盐胁迫,通过测定生长指标及叶绿素含量、可溶性糖含量、可溶性蛋白含量、相对电导率、丙二醛(MDA)含量、过氧化物酶(POD)、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性共8个生理指标来分析其综合生理抗逆性。结果表明,南方地区的5种胁迫皆对紫花苜蓿的生长生理造成了显著的抑制作用(P<0.05),其中高温、低温及渍水胁迫下,WL712的生长情况相对其他两种胁迫更差,其株高、根长和生物量受到严重抑制,渗透调节物质(可溶性糖和可溶性蛋白)在胁迫下的提升效果相对较弱;其叶绿素含量的积累则在低温和渍水胁迫下受到严重抑制,叶片和根系中3种抗氧化酶(POD、SOD和CAT)对活性氧的清除能力相对较弱。综上所述,WL712自身具有相对较差的耐热性、耐寒性以及耐渍水能力,其中对低温和渍水尤其敏感,因此在南方地区进行种植时,应尽量避开低温潮湿和高热多雨的地区,或在种植前预处理并做好相应的管理措施,以保证该品种的产量和品质。
Alfalfa(Medicago sativa L.)WL712,originally from the United States,has been introduced and cultivated in Southern China due to its high yield and premium quality.However,it exhibits limited tolerance to specific abiotic stresses.The objective of this study was to investigate the growth and physiological characteristics of this variety under various stresses,including high temperature,low temperature,drought,waterlogging,and salt stress.The overall physiological stress resistance was evaluated by measuring growth and eight physiological indices,including chlorophyll content,soluble sugar content,soluble protein content,Relative electrical conductivity(REC),malondialdehyde(MDA)content,and the activities of three antioxidant enzymes(superoxide dismutase,SOD;catalase,CAT;peroxidase,POD).The results showed that the growth of alfalfa was significantly inhibited by all five stresses(P<0.05).Among them,high temperature,low temperature,and waterlogging stress exerted a more pronounced inhibitory effect on the growth of WL712 compared to the other two kind of stresses.The height,root length,and biomass of alfalfa were substantially decreased,while the contents of osmoregulation substances(soluble sugar and soluble protein)were relatively limited.Under low temperature and waterlogging stress,the chlorophyll content significantly decreased,and the three antioxidant enzymes(POD,SOD,and CAT)in leaves and roots,responsible for eliminating reactive oxygen species,were relatively low.In summary,WL712 exhibits limited resistance to heat,cold,and waterlogging,particularly being vulnerable to low temperatures and waterlogging.Therefore,it is advisable to avoid planting WL712 in damp and rainy areas or regions with extreme temperatures in the southern region.Appropriate management practices are necessary to ensure optimal yield and quality.
