离子液体辅助药物经皮递送的研究进展

摘要 经皮给药系统具有患者依从性好、持续且受控给药、较高的局部浓度和避免新陈代谢等优势,是临床用药的有效途径之一。然而,因为人体皮肤具有多层屏障性质,大多数难溶性和大分子药物的透皮生物利用度较低,导致治疗效果不理想。近年来,离子液体(ionic liquids,ILs)的不断发展使这些药物透皮给药成为可能。ILs在室温下呈液态,是由阴离子和阳离子相互组成,但传统ILs的毒性和不稳定性限制了其应用。本文综述了皮肤结构、适用于经皮递送的ILs阴阳离子种类及其优缺点、药物经皮吸收的途径,及ILs促进药物透皮吸收的机制,分类介绍了影响ILs促进药物皮肤吸收的因素,最后对ILs促进药物透皮进行了展望。 Transdermal drug delivery system has the advantages of good patient compliance,continuous and controlled administration,high local concentration and avoidance of metabolism.Therefore,it is one of the effective ways of clinical medication.However,due to the multi-layer barrier properties of human skin,the transdermal bioavailability of most poorly soluble and macromolecular drugs is low,resulting in unsatisfactory therapeutic effects.In recent years,the continuous development of ionic liquids(ILs)has made it possible for these drugs to be transdermally administered.ILs are generally liquid at room temperature,which are composed of anions and cations,but the toxicity and instability of traditional ILs limit their application.This paper reviewed the skin structure,the types,the advantages and disadvantages of ILs anions and cations suitable for transdermal delivery,the ways of drug transdermal absorption,and the mechanism of promoting drug transdermal absorption by ILs.The factors affecting the promotion of drug transdermal absorption by ILs were introduced in categories.Finally,the prospect of promoting drug transdermal absorption by ILs was prospected.
