水杨梅总黄酮改善衰老小鼠认知功能作用机制研究 被引量:2

摘要 目的:探讨水杨梅总黄酮(ZHT)改善衰老小鼠认知功能的作用机制。方法:(1)60只雄性小鼠,随机分为正常对照组、模型组、阳性对照组、ZHT高、中、低剂量组,每组10只。空白对照组小鼠每日于颈背处皮下注射0.9%生理盐水,模型组、阳性对照组、ZHT高、中、低剂量组小鼠每日于颈背处皮下注射300 mg/kg的D-半乳糖。模型组、阳性对照组每日D-半乳糖造模后灌胃生理盐水,ZHT高、中、低剂量组每日D-半乳糖造模后分别灌胃ZHT 92 mg/100 g、46 mg/100 g、23 mg/100 g,阳性对照组灌胃茴拉西坦片4 mg/100 g,实验进行8 W。(2)采用Morris水迷宫试验、避暗试验、跳台试验观察小鼠行为学变化;采取苏木精-伊红(HE)染色观察各给药剂量组,以及模型小鼠的海马组织形态、结构的变化;采用酶标仪按试剂盒说明书检测小鼠海马组织匀浆超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的活性及丙二醛(MDA)的含量;实时荧光定量RT-PCR法检测各给药剂量组,以及模型小鼠海马组织中BDNF和TrkB mRNA的表达。结果:与正常组相比,模型组认知功能下降(P<0.05),与模型组相比,ZHT给药组认知功能有所提高(P<0.05);HE染色显示模型组小鼠海马神经细胞失去了有规律的轮廓,排列紊乱、稀疏。给予ZHT后海马神经元排列较整齐,变性神经元减少;细胞形态完整,胞核与胞浆界限清晰;酶标仪检测ZHT给药组SOD、GSH-Px活性与模型组相比显著提高(P<0.05),给药组MDA含量与模型组相比显著下降(P<0.05);与正常对照组相比,模型组SOD、GSH-Px活性显著下降(P<0.05),MDA含量显著升高(P<0.05);ZHT给药组海马组织BDNF和TrkB mRNA的表达量与模型组比较明显升高,模型组与正常组相比明显降低(P<0.05)。结论:ZHT有一定的抗衰老减轻认知障碍作用,其发挥作用可能是通过其抗氧化应激反应,上调海马组织中BDNF和TrkB mRNA的表达实现。 Objective:To investigate the mechanism of total flavone of waxberry(ZHT)in improving cognitive function in aged mice.Methods:①After adaptive feeding for 1W,60 male mice were randomly divided into a normal control group,model group,positive control group,ZHT high-dose,medium-dose,and low-dose groups,with 10 mice in each group.Mice in the blank control group were subcutaneously injected with 0.9%normal saline daily at the nape of the neck,while mice in the model group,positive control group,ZHT high-dose,medium-dose,and low-dose groups were subcutaneously injected with 300 mg/kg D-galactose daily at the nape of the neck.The model group and positive control group were given normal saline every day after D-galactose modeling.ZHT 92 mg/100 g,46 mg/100 g,23 mg/100 g after D-galactose modeling were given to the high-dose,medium-dose,and low-dose ZHT groups,respectively.Aniracetan tablets 4 mg/100 g were given to the positive control group.②Morris water maze test was used to investigate the behavior changes of mice avoiding darkness and jumping platform.Hematoxylin and eosin(HE)staining was used to observe the changes in hippocampal morphology and structure in each dose group and model mice.The activities of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)and the content of malondialdehyde(MDA)in mouse hippocampal homogenate were detected by microplate plate according to the kit instruction.The expressions of BDNF and TrkB mRNA in hippocampus of each dose group and model mice were detected by real-time fluorescent quantitative RT-PCR.Results:Compared with the normal group,the cognitive function of the model group was decreased(P<0.05),compared with the model group,the cognitive function of the ZHT group was improved(P<0.05).HE stains showed that hippocampal neurons in the model group lost regular contour.Disordered and sparse.After ZHT administration,hippocampal neurons were arranged neatly and denatured neurons were reduced.The cell morphology was intact,and the boundary between the nucleus and cytoplasm
