响应面法优化提取绵竹叶竹蜡及其成分分析

Optimization of wax extraction from Bambusa intermedia Hsueh et Yi leaves by response surface methodology and analysis of its component

摘要 利用响应面优化法提取绵竹叶的竹蜡,设定料液比、提取温度和时间三水平因素,在单因素的基础上,优化提取工艺,得到最佳工艺为:料液比1∶24 g/mL、提取时间40 min、提取温度71℃,3次平行试验绵竹叶竹蜡平均提取率为0.0223%。采用GC-MS进行成分分析,共检出37种化合物,鉴定出23种化合物。其中包含烷烃类、醇类、酯类、芳烃类、烯烃类、萘的衍生物等多种化合物,主要成分为正三十一烷(42.70%)、24-亚甲基胆固醇(14.89%)、正十四烷(7.33%)和1,1,1,2,2-五氟化丙酸三十八酯(6.25%),其中正三十一烷、24-亚甲基胆固醇具有很好的生物活性。 Response surface methodology was used to extract bamboo wax from Bambusa intermedia Hsueh et Yi leaves.material liquid ratio,extraction temperature and time were chosen as the horizontal factors.On the basis of single factor experiments,the extraction process of bamboo wax from Bambusa intermedia Hsueh et Yi leaves was optimized.The optimal extraction conditions were as follows:material to liquid ratio 1∶24 g/mL,extraction time 40 min and extraction temperature 71℃.The average extraction rate of bamboo wax from Bambusa intermedia Hsueh et Yi leaves was 0.0223%in three parallel tests.A total of 37 compounds were detected and 23 compounds were identified by GC-MS.Alkanes,alcohols,esters,aromatic hydrocarbons,olefins,naphthalene derivatives were detected.The main components were n-dodecane(42.70%),24 methylene cholesterol(14.89%),n-tetradecane(7.33%)and 1,1,1,2,2-trifluoropropionic acid octadecyl ester(6.25%).Among them,n-triacontane and 24 methylene cholesterol had good biological activities.
