摘要
目的探究气相色谱-质谱(GC-MS)和LC-MS/MS分析苏合香有效成分及药代动力学,在找出有效部位中的化学成分及其共性的同时研究其主要有效成分的药代动力学。方法应用溶剂提取对苏合香中石油醚、乙醚中有效成分进行分析,通过GC-MS联用技术,对予以化学成分的鉴定。LC-MS/MS分析对苏合香中肉桂酸药代动力学研究考察样品制备方法和取血时间,并对大鼠血药浓度予以检测,绘制血药浓度-时间曲线图,并获取药代动力学相关参数。结果经鉴定苏合香石油醚23个化合物,乙醚部位共存在11个化合物,肉桂酸的线性范围均为5~1000μg·L^(-1),方法学考察符合实验要求。日内以及日间变异系数(RSD)低于15%,精密度以及准确度均符合生物样品分析要求。肉桂酸口服给药后的Tmax、Cmax、AUC0-t、衰期t1/2及平均驻留时间MRT分别为4 h、849.29μg·L^(-1)、10577.38μg·L^(-1)·h、5.20 h、9.50 h。结论获得有效部位化学成分,研究发现GC-MS和LC-MS/MS分析应用于有效成分及药代动力学的检测准确灵敏,并适于对大鼠口服苏合香后的肉桂酸的药代动力学研究,GC-MS和LC-MS/MS分析可以作为适宜临床应用的药代动力学检测手段。
Objective To explore the active components and pharmacokinetics of Suhexiang(storax)by gas chromatography-mass spectrometry(GC-MS)and LC-MS/MS and find out the chemical components and their common characteristics in the ef-fective parts and study the pharmacokinetics of the main active components.Methods The active components of petroleum ether and diethyl ether were analyzed by solvent extraction,and the chemical components were identified by GC-MS.LC-MS/MS a-nalysis was used for pharmacokinetic study of cinnamic acid in Suhexiang(storax),the sample preparation method and blood ex-traction time were investigated,and the blood concentration of rats was detected,and the blood concentration time curve was drawn,and the pharmacokinetic parameters were obtained.Results A total of 23 compounds and 11 compounds were identified from petroleum ether extract and diethyl ether extract.The linear range of cinnamic acid was 5-1000μg·L^(-1),and the methodo-logical investigation met the experimental requirements.The intra-day and inter-day coefficient of variation(RSD)was less than 15%,and the precision and accuracy met the requirements of biological sample analysis.After oral administration of cinnam-ic acid,the Tmax,Cmax,AUC0-T,T1/2 of decay period and average dwell time MRT were 4 h,849.29μg·L^(-1),10577.38μg·L^(-1)·h,5.20 h and 9.50 h,respectively.Conclusion The chemical components of the active parts were ob-tained.It was found that GC-MS and LC-MS/MS analysis were accurate and sensitive for the detection of active components and pharmacokinetics,and were suitable for the pharmacokinetic study of cinnamic acid after oral administration of Suhexiang(sto-rax).GC-MS and LC-MS/MS analysis can be used as a suitable means of pharmacokinetic detection for clinical application.
