铁死亡对骨肉瘤进展作用影响的研究

摘要 骨肉瘤是一种具有高度侵袭性及转移性的恶性骨肿瘤,在青少年及儿童中发生率不少于70%,肿瘤好发部位常见于长骨的干骺端。15%~20%的患者在明确诊断时,已存在肉眼可见的转移病灶,其中最常见于肺转移[1]。骨肉瘤治疗的标准化方案已在临床上广泛应用。新辅助化疗、手术、术后放化疗及靶向治疗等序贯治疗手段改善了骨肉瘤患者的生存质量及预后。 Osteosarcoma is a kind of malignant bone tumor originating from primitive osteoblastic mesenchymal cells,which is prone to relapse and distant metastasis in children and adolescents.In the past 30 years,systematic and standardized treatment of osteosarcoma has been developed,including neoadjuvant chemotherapy,surgery and postoperative chemoradiotherapy.However,due to the limitations of traditional treatment and tumor heterogeneity,the five-year survival rate of osteosarcoma patients has not been substantially improved.Ferroptosis is a new modality of programmed cell death induced by ferrous ion overload and lipid peroxide accumulation.It shows great potential in inducing tumor cell death and reversing drug resistance.In order to seek a new treatment for osteosarcoma,research teams at home and abroad have made some achievements in the study of the mechanism of osteosarcoma and ferroptosis.With the development of molecular targeting and biological nanotechnology,targeted regulation of ferroptosis in tumor cells has been widely concerned by scholars.This article reviews the progress of ferroptosis mechanism in the treatment of osteosarcoma.
