摘要
在细胞试验中,使用不同浓度的三七总皂苷(panax notoginseng saponins,PNS)处理OGD/R诱导的bEnd.3细胞,研究三七总皂苷对脑缺血再灌注的保护作用。结果表明:PNS增强OGD/R诱导的脑内皮细胞bEnd.3活性,降低LDH表达,减轻迁移和侵袭能力损伤,提高ZO-1、claudin-5、occludin表达,增加细胞形成的管腔数量,激活PPARα/Nrf2信号。该研究为PNS在减轻OGD/R诱导的内皮细胞屏障和功能损伤中的应用提供了理论依据。
In cellular experiments,varying concentrations of panax notoginseng saponins(PNS)was used to treat OGD/R-induced bEnd.3 cells so as to investigate the protective effects of PNS on cerebral ischemia reperfusion.The results showed that PNS could enhance cell viability,reduce LDH level,alleviate migration and invasion ability damage,elevate ZO-1,claudin-5 and occluding levels,increase lumen number and activate PPARα/Nrf2 signal in OGD/R-induced bEnd.3 cells.This study provides a theoretical basis for the application of PNS in the alleviation of OGD/R-induced endothelial cell barrier and functional impairment.
