摘要
目的:探讨壮药金不换对脂多糖(Lipopolysaccharides,LPS)诱导的小鼠单核巨噬细胞白血病细胞(RAW264.7)炎症反应的作用。方法:观察加入脂多糖24 h后金不换提取物对Raw264.7细胞形态的影响,采用MTT法检测壮药金不换在无脂多糖刺激和加入脂多糖刺激条件下RAW264.7细胞活力,采用Griess法检测细胞上清液中一氧化氮(NO)释放量,采用酶联免疫吸附试验(Elisa)检测RAW264.7细胞上清液中炎症因子IL-6、IL-1β和TNF-α的含量。结果:0.1875~3.0μg/mL的金不换提取液在无脂多糖刺激和加入脂多糖刺激条件均不影响细胞活力(p>0.05),金不换能改善脂多糖刺激RAW264.7细胞形态,显著降低释放NO水平,同时降低炎症因子IL-6、IL-1β和TNF-α的水平。结论:金不换可通过抑制NO释放和炎症因子水平发挥抗炎作用。
Objective:To investigate the effects of Stephania on lipopolysaccharide(LPS)-induced inflammation in RAW264.7 cells.Methods:The morphology of Raw264.7 cells were observed after adding LPS for 24 h.The cell viability of RAW264.7 cells without LPS stimulation and with LPS stimulation was detected by MTT method.The release of NO in the cell supernatant was detected by Griess method.And the contents of inflammatory cytokines IL-6,IL-1βand TNF-αin the cell supernatant were detected by enzyme-linked immunosorbent assay(Elisa).Results:There is no significant impact on RAW264.7 cells viability at concentrations of 0.1875~3.0μg/mL Stephania(p>0.05).Stephania improve the cell morphology of RAW264.7 stimulated by LPS,significantly reduce the level of NO released,and significantly reduce the levels of inflammatory cytokines IL-6,IL-1βand TNF-α.Conclusion:Stephania could play anti-inflammatory role by inhibit NO release and inflammatory cytokine levels.
