二甲双胍影响恶性肿瘤免疫逃逸的研究进展

摘要 免疫逃逸是恶性肿瘤形成和治疗失败的重要原因,二甲双胍具有调节免疫和抗肿瘤作用。近年来,二甲双胍在预防肿瘤免疫逃逸中发挥积极作用,其通过抑制肿瘤细胞诱导的M2型巨噬细胞极化、调节主要组织相容性复合体分子异常表达、抑制共抑制分子表达、拮抗骨髓来源抑制性细胞和调节性T细胞的生成、抑制肿瘤细胞的抗凋亡作用等多个肿瘤免疫逃逸途径发挥作用。二甲双胍单独使用或联合化疗及免疫治疗在一定程度上提高了机体的肿瘤清除能力和免疫活性,显示出良好的抑制肿瘤免疫逃逸和抗肿瘤作用,未来可对其机制进行深入研究。 Immune escape is an important reason for tumor formation and treatment failure,and metformin has immunomodulatory and anti-tumor effects.In recent years,metformin plays an active role in preventing tumor immune escape by inhibiting M2 macrophage polarization induced by tumor cells,regulating the abnormal expression of major histocompatibility complex molecules,inhibiting the expression of co-inhibitory molecules,and inhibiting the generation of bone marrow derived suppressor cells and regulatory T cells,inhibiting the anti-apoptosis of tumor cells and other tumor immune escape pathways.The body′s capability of antitumor and immunological competence are improved by metformin monotherapy or its combination with chemotherapy/immunotherapy.Metformin displays good inhibitory effects of tumor immune escape and anti-tumor effects,and further research can be conducted on the mechanisms in the future.
