摘要
【目的】花椒黄花病(flower yellowing disease,FYD)是近年来花椒重要病害之一,可导致感病花椒减产,本研究应用高通量测序技术分析花椒黄花病潜在病原,为该病害病原种类鉴定、病害防治提供理论依据。【方法】取竹叶花椒永青1号的黄花病植株4份和无症植株2份的花、叶样品进行病毒组测序分析。【结果】测序数据分析发现,6个样品共有484个contigs与病毒序列相似,分别匹配到12个病毒科的15种病毒。6个样品均检测出在九叶青花椒上已报道的4种病毒:四川青花椒线虫多面体病毒(GSP⁃NeV)、四川青花椒悬钩子病毒(GSPIV)、四川青花椒耳突花叶病毒(GSPEV)、四川花椒弹状病毒(GSPNuV)。发现花椒中未报道的2种新病毒,分别命名为永青花椒相关幽影病毒(Yongqing green pepper⁃associated umbravirus)与永青花椒相关负链RNA病毒(Yongqing green pepper⁃associated ssRNA negative⁃strand virus);此外,在感病D⁃2和无症H⁃1样品中发现马铃薯Y病毒(Potato virus Y,PVY)。从感病样品D⁃2、D⁃3、D⁃4发现乙型线形病毒科(Betaflexiviridae)香石竹潜隐病毒属(Carlavirus)马铃薯M病毒(Potato virus M,PVM)、马铃薯H病毒(Potato virus H,PVH)和马铃薯S病毒(Potato virus S,PVS),在D⁃1花椒发现葡萄茎痘相关病毒(Gra⁃pevine rupestris stem pitting⁃associated virus,GRSPAV)。【结论】从永青1号花椒测序样品中均发现已报道的4种病毒以及未报道的2种新病毒,并检测出仅存在于感病组的香石竹潜隐病毒属,推测高表达的香石竹潜隐病毒属病毒或可视为诱发花椒黄花病的因素之一。
【Objective】Flower yellowing disease(FYD)is one of the most significant diseases affecting Zanthoxylum sp.in recent years,leading to yield losses in susceptible Zanthoxylum spp.In the study,high⁃throughput sequencing technology was employed to analyze the po⁃tential pathogens responsible for FYD,aiming to provide a basis for pathogen identification and disease control.【Method】The flower and leaf samples of four plants with FYD symptoms and two asymptomatic plants of Z.armatum var.yongqing 1 were collected for virome analy⁃sis.【Result】Sequencing data analysis indicated that a total of 484 contings from six samples matrched 15 virus species belonging to 12 differ⁃ent viral families.Among these six samples,four previously reported viruses were detected in Z.armatum var.novemfolius:Green Sichuan pepper⁃nepovirus(GSPNeV),Green Sichuan pepper⁃idaeovirus(GSPIV),Green Sichuan pepper⁃enamovirus(GSPEV),Green Sichuan pepper⁃nucleorhabdovirus(GSPNuV).Notably,previously unreported viruses in Zanthoxylum spp.included two novel RNA viruses tenta⁃tively named Yongqing green pepper⁃associated umbravirus and Yongqing green pepper⁃associated ssRNA negative⁃strand virus.Additional⁃ly,Potato virus Y(PVY)was detected in symptomatic D⁃2 and asymptomatic H⁃1 samples.Potato virus M(PVM),Potato virus H(PVH),Potato virus S(PVS),all belonging to the genus Carlavirus in the family Betaflexiviridae,were identified in FYD plant samples of D⁃2,D⁃3 and D⁃4.Furthermore,the grapevine rubestris stem pitting associated virus(GRSPAV)of the genus Foveavirus was detected ex⁃clusively in the D⁃1 sample.【Conclusion】All four reported viruses as well as two new unreported viruses were detected in the sequenced samples from Yongqing 1,and the genus Carlavirus,which was only present in the symptomatic group,was detected,and it was inferred that the highly expressed viruses of the genus Carlavirus should be regarded as one of the factors that induced FYD.
