数据挖掘联合网络药理学探讨苦木抗肿瘤活性研究及验证 被引量:1

Data Mining and Network Pharmacology Approach to Explore and Validate the Anti-Tumor Activity of Picrasma quassioides(D.Don)Benn.

摘要 采用数据挖掘、网络药理学和体外验证对苦木抗肿瘤作用机制的初步探讨.通过文献检索和中药系统药理学数据库建立了苦木化学成分库,并根据口服利用度和药物相似性筛选出有效成分.使用Swiss Target Prediction数据库确定了苦木的潜在作用靶点,并通过Metascape进行GO和KEGG通路富集分析.利用STRING数据库构建了蛋白互作网络,并使用Cytoscape软件进行可视化.在CTD数据库中聚焦疾病相关性,并在KEGG和CBPortal数据库中预测了抗肿瘤机制.MTT比色法实验验证了苦木对多种癌细胞系的抑制作用.苦木中共检索得到53种化学成分,筛选出5种主要活性成分,并确定了309个潜在作用靶点.KEGG通路分析揭示这些靶点主要涉及癌症相关的信号途径.网络分析表明苦木可能对子宫癌、非小细胞肺癌和食道癌有影响.体外实验验证了苦木对几种癌细胞系显示出浓度依赖性的抑制作用. This paper presents a preliminary investigation of the anti-tumor mechanism of action of Picrasma quassioides(D.Don)Benn.using data mining,network pharmacology and in vitro valida-tion.A library of chemical components of Picrasma quassioides(D.Don)Benn.was established using literature search and the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).Based on the principles of oral bioavailability and druglikeness,the active medici-nal components of Picrasma quassioides(D.Don)Benn.were screened.The Swiss Target Prediction database was searched to obtain the potential targets of Picrasma quassioides(D.Don)Benn..Gene Ontology(GO)functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis were performed using the Metascape database.A protein-protein interaction(PPI)network was constructed using the STRING database and visualized using the Cytoscape software.Dis-ease relevance was focused in the CTD database,and anti-tumor mechanisms were predicted in the KEGG and CBPortal databases.The inhibitory effects of Picrasma quassioides(D.Don)Benn.on a va-riety of cancer cell lines were verified by MTT colorimetric assay experiments.A total of 53 chemical components were retrieved from the Picrasma quassioides(D.Don)Benn.,of which five major active components were screened,and 309 potential targets were obtained.KEGG pathway analysis revealed that these targets were mainly involved in cancer-related signaling pathways.Network analysis showed that Picrasma quassioides(D.Don)Benn.may have an effect on uterine cancer,non-small cell lung cancer and esophageal cancer.In vitro experiments verified that Picrasma quassioides(D.Don)Benn.showed concentration-dependent inhibitory effects on several cancer cells lines.
