摘要
目的:探讨环状RNA circ_104939在肺腺癌组织中的临床意义。方法:采集10例肺腺癌病人的肿瘤组织和癌旁组织进行环状RNA测序分析,以肿瘤组织表达明显增高的环状RNA circ_104939作为研究对象。利用qRT-PCR检测30对肺腺癌组织与癌旁组织中circ_104939的表达量,验证测序结果。用qRT-PCR检测和分析circ_104939在肺腺癌细胞系(HCC-78、A549、KTA-7以及PC-9)和正常人支气管上皮细胞BEAS-2B中的表达差异。siRNA敲减circ_104939后,用MTT、平板划痕实验、Transwell实验、流式细胞术检测细胞周期实验分别检测肺腺癌细胞(KTA-7和PC-9)增殖、迁移、侵袭以及细胞周期的变化。利用RNA pulldown和荧光素酶报告实验验证circ_104939的可能靶标miR-191-5p。siRNA敲减circ_104939后,将肺腺癌细胞(KTA-7和PC-9)分别皮下注射到裸鼠体内构建荷瘤小鼠模型,检测移植瘤体积以及质量。结果:相比癌旁组织,肺腺癌组织中的circ_104939表达显著增高(P<0.01);circ_104939在肺腺癌细胞中的表达水平明显高于正常人支气管上皮细胞(P<0.05或P<0.01)。circ_104939敲减后,KTA-7和PC-9细胞增殖、侵袭能力、细胞周期进展、PC-9细胞迁移能力下降(P<0.05或P<0.01)。circ_104939能够海绵样吸附miR-104939。circ_104939敲减后,荷瘤小鼠肿瘤体积减小(P<0.05或P<0.01),肿瘤质量显著降低(P<0.01或P<0.001)。结论:circ_104939在肺腺癌组织中呈高表达,有望作为肺腺癌诊断和治疗的靶点。
Objective:To investigate the expression level and clinical value of circ_104939 in lung adenocarcinoma.Methods:Ten pairs of lung adenocarcinoma tumor tissues and adjacent tissues were collected and detected by circular RNA sequencing,and circ_104939 with significantly higher expression in tumor tissues was taken as the research object.The technology of qRT-PCR was used to detect the expression of circ_104939 in 30 pairs of lung adenocarcinoma tissues and adjacent tissues to verify the sequencing results.The expression of circ-104939 in lung adenocarcinoma cell lines(HCC-78,A549,KTA-7 and PC-9) and normal bronchial epithelial cells BEAS-2B were detected and analyzed by qRT-PCR.After circ_104939 was knocked down by siRNA,proliferation,migration,invasion and cell cycle changes of lung adenocarcinoma cells(KTA-7 and PC-9) were detected by MTT,plate scratches assay,Transwell assay and flow cytometry cell cycle assay.RNA pulldown and luciferase reporting assay were used to verify the possible target of circ_104939.After circ_104939 was knocked down by siRNA,lung adenocarcinoma cells(KTA-7 and PC-9) were subcutaneously injected into nude mice to construct tumor-bearing mouse models,and the volume and weight of transplanted tumor were detected.Results:The expression level of circ_104939 in lung adenocarcinoma tissues was significantly higher than that in adjacent tissues(P< 0.01).The expression level of circ_104939 in lung adenocarcinoma cells was significantly higher than that in normal bronchial epithelial cells(P<0.05 or P< 0.01).After circ_104939 knockdown,KTA-7 and PC-9 cell proliferation,invasion ability,cell cycle progression and PC-9 cell migration ability decreased(P< 0.05 or P< 0.01).Mechanistically,circ_104939 serves as a sponge for miR-191-5p.After circ_104939 knockout,tumor volume decreased(P< 0.05 or P< 0.01),and tumor weight also decreased significantly(P< 0.01 or P< 0.001) in tumor-bearing mice.Conclusion:The expression of circ_104939 is highly increased in lung adenocarcinoma tissues,which is expected to
