Determination of Methyl Selenocysteine,Selenomethionine and Selenocysteine in Lentinus edodes by Liquid Chromatography Tandem Mass Spectrometry

摘要 建立一种提取和测定香菇中甲基硒代半胱氨酸(methyl selenocysteine,MeSeCys)、硒代蛋氨酸(selenom-ethionine,SeMet)、硒代半胱氨酸(selenocysteine,Sec)含量的超高效液相色谱串联质谱方法。对比3种不同前处理方法对香菇样品中硒代氨基酸的提取效果,选Agilent ZORBAX SB-AQ型亲水色谱柱(柱长100 mm,内径3.0 mm,粒径1.8μm),1%甲酸水和甲醇流动相梯度洗脱,柱温35℃,流量0.2 mL/min。结果表明,3种提取方法中,酶解提取法提取效率最高,能较好地提取香菇中甲基硒代半胱氨酸、硒代蛋氨酸、硒代半胱氨酸,采用HPLC-MS检测3种氨基酸在质量浓度0.2~20.0 ng/mL呈现较好的线性关系,平均回收率分别为92.3%,93.0%,92.4%,相对标准偏差分别为2.71%,2.14%,1.83%。该方法简单、快速、准确,适用于香菇中甲基硒代半胱氨酸、硒代蛋氨酸、硒代半胱氨酸含量的测定。 A liquid chromatography-tandem mass spectrometry method was developed for the extraction and determination of methyl Selenocysteine,selenomethionine and Selenocysteine in Lentinus edodes.Three different pretreatment methods were used for the extraction of selenoamino acids from Lentinus edodes.The Agilent Zorbax SB-AQ hydrophilic column(100 mm in length,3.0 mm in diameter and 1.8μm in particle size)was selected,1%formic acid water and methanol mobile phase gradient elution,column temperature 35℃,flow rate 0.2 mL/min.This method could extract methyl selenocysteine,selenomethionine and selenocysteine from Lentinus edodes,and the three amino acids showed a good linear relationship in the concentration range of 0.2~20.0 ng/mL.The recovery rates were detection limit.The method was simple,rapid and accurate for the determination of methyl selenocysteine,selenomethionine and selenocysteine in Lentinus edodes.
