聚乙二醇浸种对黄瓜幼苗耐冷性的影响

摘要 以“长春密刺”黄瓜为实验材料,采用不同浓度(1%,5%,15%,25%,35%)聚乙二醇(PEG)溶液常温浸种4 h,蒸馏水作为对照(CK).浸种完成后,待黄瓜幼苗长至两叶一心期,在培养箱中低温(昼/夜温度分别为:25/15(常温对照组),20/12,15/9,10/6℃)处理3 d,测定水势大小以及可溶性蛋白质、可溶性糖、脯氨酸的质量浓度,研究了PEG浸种对黄瓜幼苗耐冷性的影响.结果表明,随着处理温度的降低,PEG浸种的黄瓜幼苗水势均呈下降趋势,其叶片中可溶性糖、可溶性蛋白质以及脯氨酸的质量浓度均呈上升趋势;在相同处理温度下,经PEG浸种后,黄瓜叶片的水势以及可溶性糖、可溶性蛋白质和脯氨酸的质量浓度均显著高于对照组,其中15%PEG浸种对提升黄瓜幼苗低温耐受性最佳.PEG浸种能明显提高黄瓜幼苗耐冷性. Cucumber variety“Changchun Mici”was used as experiment materials.The seeds of this variety was soaked for four hours at room temperature with different concentrations of polyethylene glycol(PEG)solutions(1%,5%,15%,25%,35%)and those treated with H2O were applied as control(CK).After seed soaking,the cucumber seedlings were allowed to grow to the two-leaf-one-heart stage,and then treated with low temperature(day/night temperature:25/15(ambient control group),20/12,15/9,10/6℃,respectively)in artificial climate chamber for three days.The water potential,the contents of soluble protein,soluble sugar and proline were measured to investigate the effect of PEG soaking on seedlings tolerance to cold of cucumber.The results showed that water potential of PEG-soaked cucumber seedlings decreased with temperature decreasing.The contents of soluble sugar,soluble protein and proline in the leaves of PEG-soaked cucumber seedlings all presented an increasing trend.The water potential,the contents of soluble sugars,soluble proteins and proline of the leaves of PEG-soaked cucumber seedlings were significantly higher than those of the CK under the same temperature,15%PEG was the most effective treatment to enhance cold tolerance.PEG seed soaking could significantly improve cold tolerance of cucumber seedlings and seed soaking.
