簇毛麦染色体特异分子标记的筛选及应用

摘要 簇毛麦是小麦的一个野生近缘种,抗病性和抗逆性强,蛋白质和赖氨酸含量高,是小麦遗传改良的重要基因源。为定位、转移和利用簇毛麦有益基因,通过花粉辐射,获得了一批涉及簇毛麦不同染色体且不同区段的结构变异体。为了鉴定该批材料中的簇毛麦染色体身份,选用小麦EST-STS引物和小麦、大麦及黑麦SSR引物共1576对,其中755对引物在普通小麦中国春与簇毛麦间存在多态性;进一步对亲本及一套簇毛麦二体异附加系进行PCR扩增分析,Xbarc210_(-220)等2个标记可追踪簇毛麦1V染色体,Xcinau186_(-600)等14个标记可追踪簇毛麦2V染色体,Xcinau 200_(-245)等8个标记可追踪簇毛麦3V染色体,Xcinau 206_(-400)等8个标记可追踪簇毛麦4V染色体,Xcinau 211_(-700)等41个标记可追踪簇毛麦5V染色体,Xcinau 250_(-120)等2个标记可追踪簇毛麦6V染色体,Xcinau 255_(-700)等14个标记可追踪簇毛麦7V染色体。利用这些簇毛麦染色体特异分子标记鉴定辐射诱导材料的回交后代,有70个结构变异体的簇毛麦身份得到确定,表明这些标记可以用来快速检测普通小麦背景中的簇毛麦染色体或染色体片段。 Haynaldia villosa(L.)is a wild relative species of wheat(Triticum aestivum L.).It has strong ability of diseases and stress resistance,high protein and lysine contents.These characters make it an important gene pool for wheat genetic improvement.To localize,transfer and use interested genes of H.villosa,a series of involving different H.villosa chromosome and chromosomal region lines induced by irradiation.To identify H.villosa chromosomes or chromosome segments in these materials,1576 primer pairs were used including EST-STS of wheat and SSR of wheat,barley and rye,and 755 of them could amplify specific polymorphic bands between common wheat cv.Chinese Spring and H.villosa.These 755 primer pairs were further used for screening markers specific to individual chromosomes of H.villosa using 1V to 7V disomic additional lines and their parents.The results showed that 2 markers such as Xbarc210_(-220)could be used to trace chromosome 1V,14 markers such as Xcinau 186-600 to 2V,8 markers such as Xcinau 200_(-245)to 3V,8 markers such as Xcinau 206_(-400)to 4V,41 markers such as Xcinau 211_(-700)to 5V,2 markers such as Xcinau 250_(-120)to 6V,14 markers such as Xcinau 255_(-700)to 7V.These specific markers were used to identify the H.villosa chromosome and chromosome segments of backcrossed generations derived from pollen irradiation.70 structural changes involved in different H.villosa chromosomes were identified.Therefore,these chromosome-specific markers could be used to detect chromosomes and chromosome segments of H.villosa in common wheat background.
