胸腺肽α1对铜绿假单胞菌感染支气管扩张症大鼠的免疫调节作用

摘要 目的分析胸腺肽α1对铜绿假单胞菌感染支气管扩张症大鼠的免疫调节作用。方法将2022年10月至2023年2月采购的50只铜绿假单胞菌介导的Balb/c大鼠纳入研究,随机抽取10只大鼠依照常规进行组织病理学检查,均成功建模;20只大鼠接受胸腺肽α1皮下注射治疗为观察组;另20只大鼠接受生理盐水皮下注射为对照组。收集两组大鼠的支气管肺泡灌洗液与外周血为检测标本,经逆转录-聚合酶链反应(RT-PCR)检测线粒体DNA(mtDNA)含量;免疫蛋白印迹试验测定Toll样受体(TLR)9;以流式细胞术测定辅助性T细胞(Th)17细胞计数、调节性T细胞(Treg)细胞计数、白细胞分化抗原(CD)4^(+)、CD8^(+)细胞计数;以酶联免疫吸附试验测定白细胞介素(IL)-2、IL-6、IL-10、IL-17水平;综合评估胸腺肽α1对铜绿假单胞菌感染支气管扩张症免疫调节的作用。结果观察组CD4^(+)、CD8^(+)细胞计数和mtDNA、Th17、IL-10水平高于对照组,差异有统计学意义(P<0.05)。观察组TLR9、Treg、IL-2、IL-6、IL-17水平低于对照组,差异有统计学意义(P<0.05)。结论铜绿假单胞菌感染支气管扩张症大鼠的免疫力低,而胸腺肽α1可通过调节大鼠免疫功能来改善大鼠病情。 Objective To analyze the immunomodulatory effect of thymosin-α1 on rats with bronchiectasis infected by pseudomonas aeruginosa.Methods A total of 50 Balb/c rats mediated by pseudomonas aeruginosa purchased by our research team from October 2022 to February 2023 were included in the study,and 10 rats were randomly selected for histopathological examination according to routine procedures,all of which were successfully modeled.20 rats receiving a subcutaneous injection of thymosin-α1 were included in the observation group.Another 20 rats receiving a subcutaneous injection of normal saline were included in the control group.Bronchoalveolar lavage fluid(BALF)and peripheral blood were collected from the two groups of rats as test samples,and mitochondrial DNA(mtDNA)content was detected by reverse transcription-polymerase chain reaction(RT-PCR).Immunoblotting assay was used to determine Toll-like receptors(TLRs)9.Helper T cell(Th)17 cell count,regulatory T cell(Treg)cell count,Cluster of Differentiation(CD)4^(+),and CD8^(+)cell count were measured by flow cytometry.The levels of interleukin(IL)-2,IL-6,IL-10,and IL-17 were measured using enzyme-linked immunosorbent assay.By virtue of the above-mentioned methods,a comprehensive evaluation of immunomodulation of thymosin-α1 on bronchiectasis infected by pseudomonas aeruginosa was conducted.Results The CD4^(+)cell count,CD8^(+)cell count,mtDNA level,Th17 level,and IL-10 level in the observation group were higher than those in the control group,with statistically significant differences(P<0.05).The levels of TLR9,Treg,IL-2,IL-6,and IL-17 in the observation group were lower than those in the control group,with statistically significant differences(P<0.05).Conclusion The immunity of rats with bronchiectasis infected by pseudomonas aeruginosa is low,and thymosin-α1 can improve the condition of rats by regulating their immune functions.
