基于数据挖掘分析肝细胞癌中ATP柠檬酸裂解酶表达的临床意义

摘要 目的多数据库分析ATP柠檬酸裂解酶(ACLY)在肝细胞癌中的表达和临床意义。方法采用UALCAN、GEPIA和HPA数据库分析ACLY mRNA和蛋白在肝癌组织中的表达水平,ACLY异常表达与肝细胞癌患者临床病理指标和生存期的关系;采用STRING数据库分析以ACLY为中心相关蛋白之间的相互作用。结果ACLY在肝癌组织中表达水平高于正常组织,差异有统计学意义(P<0.05);肝癌组织中ACLY高表达与肿瘤临床分期、病理分级、淋巴结转移有关;癌组织高表达ACLY的肝细胞癌患者生存率低于ACLY低表达患者,差异有统计学意义(P<0.05)。ACLY及其相互作用蛋白质主要参与脂肪酸合成代谢、脂质和胆固醇代谢过程的调节。结论ACLY在肝癌组织中高表达,与肝细胞癌肿瘤生物学行为和肝细胞癌患者不良预后有关。 Objective Multi-database analysis of the expression and clinical significance of ATP-citrate lyase(ACLY)in hepatocellular carcinoma was conducted.Methods UALCAN,GEPIA and HPA databases were used to analyze the expression levels of ACLY mRNA and protein in cancer tissue,and the relationship between abnormal expression of ACLY and clinical pathological indicators and survival in hepatocellular carcinoma patients.The String database was used to analyze the interactions between related proteins centered around ACLY.Results The expression of ACLY in liver cancer tissue was higher than that in normal tissue,with statistically significant difference(P<0.05),and the high expression of ACLY was related to clinical stage,pathological grading,and lymph node metastasis of the tumor.The survival rate of hepatocellular carcinoma patients with high expression of ACLY in cancer tissue was lower than that of patients with low expression of ACLY,with statistically significant difference(P<0.05).ACLY and its interacting proteins were mainly involved in the regulation of fatty acid synthesis metabolism,and lipid and cholesterol metabolism.Conclusion ACLY is highly expressed in liver cancer tissue and is associated with the biological behavior of hepatocellular carcinoma and poor prognosis in hepatocellular carcinoma patients.
