摘要
目的:建立高效液相色谱法(HPLC)测定滇产白花蛇舌草中去乙酰车叶草酸甲酯的含量,提高白花蛇舌草定性定量的专属性鉴别。方法:采用Zorbax Eclipse XDB-C 18(4.6 mm×250 mm,5μm)色谱柱;流动相∶乙腈-水(3.5∶96.5);检测波长:238 nm;柱温:25℃;流速:1.0 mL/min;进样量:10μL。结果:白花蛇舌草检出去乙酰车叶草酸甲酯,其伪品未检出,乙酰车叶草酸甲酯在3.7683~241.1684μg/mL(r=0.99985)范围内线性关系良好;平均回收率为94.40%,RSD为2.3%。结论:建立的方法,可用于白花蛇舌草与伪品的鉴别,为评价该药材质量具有借鉴意义。
Objective To determinethe content of deacetylated methyloxalate in Hedyotis diffusa produced in yunnan by HPLC-DAD,to improve qualitative method of Hedyotis diffusa.Methods The Zorbax Eclipse XDB-C 18 column(4.6 mm×250 mm,5μm)was used for determination ofdeacetylated methyloxalate.Gradient elution with acetonitrile and water was performed(the proportion is 3.5∶96.5).The deacetylated methyloxalate was detected at a wavelength of 238 nm;the system operated at 25℃;flow rate was 1 mL/min and the injection volume was 10μL.Results The deacetylated methyloxalate was detected in Hedyotis diffusa,but the counterfeit was not detected.The deacetylated methyloxalateshowed good linearity in the ranges of 3.7683-241.1684μg/mL(r=0.99985),and the average recoveries were 94.40%,RSD=2.3%.Conclusion The HPLC method established is a rapid and accuracy,and can be used to distinguish Hedyotis diffusa from counterfeit,andis helpful to tis quality control.
