摘要
基于QuEChERS的快速、简便的优势,建立了一种同时测定食品中展青霉素(PAT)、呕吐毒素(DON)、黄曲霉毒素B_(1)、B_(2)、G_(1)、G_(2)(AFB_(1)、AFB_(2)、AFG_(1)、AFG_(2))、玉米赤霉烯酮(ZEN)、赭曲霉毒素A(OTA)等8种真菌毒素的方法。样品采用乙腈-水-乙酸(70:29:1,V/V/V)混合提取液提取,经乙二胺-N-丙基硅烷(PSA)、碳十八键合硅胶(C18)、石墨化碳黑(GCB)和无水硫酸镁混合型固相分散净化,净化液过滤膜后通过Phenomenex Luna C_(18)(2)100Å(150×2.0 mm,3μm)色谱柱分离,以电喷雾电离串联质谱多反应监测模式(MRM)进行确证和检测,氘代同位素内标法定量,分别对流动相、提取溶剂、固相净化剂的选择进行了优化。结果表明:8种真菌毒素显示了良好的线性关系(r≥0.9984);在低、中、高3个添加水平下目标物的加标平均回收率为70.1%~119%,相对标准偏差(RSD)为2.45%~9.97%;方法的检出限(LOD,S/N=3)为0.28~42.18μg/kg。该方法前处理快速简便、灵敏度高、重现性好,适用于花生、玉米、薏米仁中多种真菌毒素的测定。
Based on the quick and easy advantages of QuEChERS,a method was developed for the simultaneous determination of patulin(PAT),emetic toxin(DON),aflatoxins AFT(AFB1,AFB2,AFG1,AFG2),zearalenone(ZEN)and Ochratoxin a(OTA)in foods.The samples were extracted with acetonitrile-water-acetic acid(70:29:1,V/v/V)mixture,after purification by solid dispersion of ethylenediamine-n-propyl silane(PSA),C_(18) bonded silica gel(C_(18)),graphitized carbon black(GCB)and anhydrous magnesium sulfate,the extract was separated on a Phenomenex Luna C_(18)(2)100Å(150×2.0 mm,3μm)column,confirmed and detected by electrospray ionization tandem mass spectrometry multiple reaction monitoring(MRM),and quantified by deuterium isotope internal standard method.The choice of mobile phase,extraction solvent and solid-phase purifying agent was optimized.The results showed that the 8 mycotoxins showed a good linear relationship(r≥0.9984),and the average recoveries of the target compounds were 70.1%~119%at low,middle and high levels,the relative standard deviation(RSD)was 2.45%~9.97%,and the detection limits(LOD,S/N=3)were 0.28~42.18μg/kg,respectively.The method is rapid,simple,sensitive and reproducible,and is suitable for the determination of various mycotoxins in corn,peanut and edible oil.
