摘要
为进一步探究胸膜肺炎放线杆菌(Actinobacillus pleuropneumoniae,APP)感染仔猪的致病机制,利用SYBR Green荧光定量PCR方法,探究仔猪感染后IL-1β、IL-6、IL-8及caspase-3基因表达水平的变化。通过肺灌洗液获得猪肺巨噬细胞,将APP以MOI=10的剂量感染细胞,并对经APP感染后的肺巨噬细胞相关细胞因子(IL-1β、IL-6、IL-8及caspase-3)基因表达水平进行检测。结果显示,APP感染后3~12 h,IL-1β、IL-6、IL-8及caspase-3基因表达量呈升高趋势,感染后12~24 h达到高峰。结果提示,APP感染早期IL-1β、IL-6、IL-8等细胞因子在宿主清除致病菌过程中发挥着重要作用。本研究为下一步深入探索APP致病机制奠定了基础。
In order to further investigate the pathogenic mechanism of Actinobacillus pleuropneumoniae(APP)infection in piglets,the changes in the expression levels of IL-1β,IL-6,IL-8 and caspase-3 genes in infected piglets were investigated by SYBR green fluorescence quantitative PCR assay.Porcine alveolar macrophages(PAMs)were obtained through bronchoalveolar lavage fluid and infected by APP at a dose of MOI=10,then the expression levels of relevant cytokine genes(IL-1β,IL-6,IL-8 and caspase-3)were detected.The results showed that the amounts of gene expression of IL-1β,IL-6,IL-8 and caspase-3 were increasing at 3~12 h after APP infection,and reached a peak at 12~24 h.In conclusion,IL-1β,IL-6 and IL-8 played an important role in clearing pathogenic bacteria by hosts at early stage of APP infection.A foundation was laid for further exploration of APP's pathogenic mechanism.
