基于超高效液相色谱-质谱联用法的过敏煎、加味过敏煎大鼠血清差异代谢物分析 被引量:1

Study on Serum Differential Metabolites in Rats After Oral Administration of Guominjian Decoction and Modified Guominjian Decoction by UHPLC-QE-MS

摘要 目的基于非靶代谢组学研究过敏煎、加味过敏煎中药复方灌胃给药后大鼠血清中两复方的主要差异代谢物,为进一步研究加味过敏煎缓解瘙痒机制提供基础。方法基于超高效液相色谱-质谱联用技术,通过对过敏煎含药血清、加味过敏煎含药血清进行主成分分析、正交偏最小二乘法-判别分析等方法进行数据模式识别,并根据差异倍数>1.2或<0.83,变量投影重要度>1且P<0.05为筛选标准,筛选出差异代谢物;利用MetaboAnalyst 5.0对差异物进行通路分析及富集分析。结果共筛出173个差异代谢物;通路分析得出2条相关通路:苯丙氨酸、酪氨酸和色氨酸的生物合成通路及维生素B6代谢通路;富集到2个差异代谢物:酪氨酸和吡哆醇。结论过敏煎、加味过敏煎含药血清最主要的血清差异代谢物可能为酪氨酸及吡哆醇,加味过敏煎的瘙痒治疗优势,可能跟酪氨酸、吡哆醇有关。 Objective To study the main differential metabolites in rats serum after intragastric administration of Guominjian Decoction and Modified Guominjian Decoction based on non-targeted metabolomics technology,so as to provide the basis for further research on the mechanism of Modified Guominjian Decoction in relieving pruritus.Methods Based on ultra high performance liquid chromatography-quadrupole electrostatic field orbital trap-mass spectrometry(UHPLC-QE-MS)non-targeted metabolomics technology,the data pattern recognition was carried out by analyzing the drug-containing serum of Guominjian Decoction and Modified Guominjian Decoction,principal component analysis(PCA),orthogonal projections to latent structures-discriminant analysis(OPLS-DA)and other analytical methods.Differential metabolites were screened according to fold change(FC)greater than 1.2 or less than 0.83,variable importance in the projection(VIP)greater than 1 and P-value less than 0.05 as screening criteria.MetaboAnalyst 5.0 was used to conduct pathway analysis and enrichment analysis for the differential metabolites.Results A total of 173 differential metabolites were screened.And after pathway analysis,two related pathways were obtained:phenylalanine,tyrosine and tryptophan biosynthesis,and vitamin B6 metabolism.There were two differential metabolites enriched in the pathways:tyrosine and pyridoxine,respectively.Conclusion Tyrosine and pyridoxine may be the main serum differential metabolites in the drug-containing serum of Guominjian Decoction and Modified Guominjian Decoction.The advantage of Modified Guominjian Decoction in the treatment of pruritus may be related to tyrosine and pyridoxine.
