干酪乳杆菌SY13对小鼠免疫功能和肠道菌群的影响

摘要 在前期已验证干酪乳杆菌SY13具有降脂益生功能的基础上,结合低聚糖可促进益生菌在肠道定殖的特性,本试验拟深入探讨SY13对宿主免疫功能和肠道菌群的影响,将48只6周龄小鼠分为4个组:分别灌服无菌PBS(BP1组)、SY13(BP2组)、SY13+低聚果糖(BP3组)和SY13+乳果糖(BP4组),连续28 d,分别于第1、3、5、7天,每组选3只小鼠眼球取血,用ELISA测定小鼠血清分泌型免疫球蛋白A(sIgA)和细胞因子(IL-1β、IL-2、IL-4和IL-10)含量;采用TaqMan-MGB探针,通过实时荧光定量PCR检测第1天各组小鼠肠道内容物中SY13含量;利用Illumina Miseq分析各组小鼠第1、7天的盲肠微生物多样性。结果显示,第7天时BP1组小鼠血清sIgA含量显著低于其他各组(P<0.05);第3天时BP4组小鼠血清IL-1β和第5天时血清IL-4、IL-10含量均显著高于其他各组(P<0.05);第1天时BP4组小鼠空肠、回肠和盲肠中SY13含量显著高于其他各组(P<0.05);第1、7天时BP3组和BP4组小鼠盲肠拟杆菌门菌群丰度显著高于BP1组(P<0.05),BP3组小鼠盲肠厚壁菌门与拟杆菌门菌群丰度比值低于其他各组;第7天时,BP3组和BP4组小鼠盲肠另枝菌属菌群丰度显著高于BP1组(P<0.05)。结果表明,SY13可促进机体健康,同时服用SY13和乳果糖可调节宿主肠黏膜免疫,改善肠道菌群,降低肠道感染概率,而同时服用SY13和低聚果糖主要是通过与肠道菌群相互作用发挥益生功效。 Building on the previously validated lipid-lowering and probiotic functions of Lactobacillus casei SY13,combined with the characteristic of oligosaccharide promoting the colonization of probiotics in the intestines,this study aims to investigate the effects of SY13 on the host's immune function and intestinal flora.Forty-eight six-week-old mice were divided into four groups:gavaged with sterile PBS(BP1 group),SY13(BP2 group),SY13+fructo-oligosaccharide(BP3 group),and SY13+lactulose(BP4 group)for 28 days.On the 1 st,3 rd,5 th,and 7 th days,blood samples were collected from three mice in each group for ELISA to determine serum secretory immunoglobulin A(sIgA)and cytokine levels(IL-1β,IL-2,IL-4 and IL-10).On the 1 st day,TaqMan-MGB probes were used for real-time quantitative PCR to detect SY13 content in the intestinal contents of mice in each group.On the 1 st and 7 th days,the microbial diversity in the cecum of mice in each group was analyzed using Illumina Miseq.The results showed that on the 7 th day,the serum sIgA level in the BP1 group was significantly lower than in the other groups(P<0.05).On the 3 rd day,the serum IL-1βlevel in the BP4 group,and on the 5 th day,the serum IL-4 and IL-10 levels in the BP4 group were significantly higher than in the other groups(P<0.05).On the 1 st day,the SY13 content in the jejunum,ileum,and cecum of the BP4 group was significantly higher than in the other groups(P<0.05).On the 1 st and 7 th days,the abundance of Bacteroidetes in the cecum of the BP3 and BP4 groups was significantly higher than in the BP1 group(P<0.05),and the ratio of Firmicutes to Bacteroidetes in the cecum of the BP3 group was lower than in the other groups.On the 7 th day,the abundance of Alistipes in the cecum of the BP3 and BP4 groups was significantly higher than in the BP1 group(P<0.05).The results indicate that SY13 promotes overall health,and concomitant use of SY13 and lactulose regulates host intestinal mucosal immunity,improves intestinal flora,reduces the risk of intestinal infections,while
