摘要
巨噬细胞在心肌梗死后的炎症和纤维化过程中发挥重要的调控作用。巨噬细胞根据有无CC类趋化因子受体2(CCR2)表达分为CCR2^(+)巨噬细胞和CCR2^(-)巨噬细胞。CCR2^(-)巨噬细胞通过Ly6C的表达水平分为促炎的Ly6C^(hi)CCR2^(hi)CX3CR1^(lo)巨噬细胞和抑炎的Ly6C^(lo)CCR2^(lo)CX3CR1^(hi)巨噬细胞。心肌梗死后的早期阶段和急性炎症期Ly6C^(hi)巨噬细胞产生炎症细胞因子,Ly6C^(lo)巨噬细胞在梗死后的后期被招募,分泌抗炎细胞因子和生长因子。巨噬细胞通过旁分泌白细胞介素(IL)-1β、IL-6和肿瘤坏死因子-α等炎症因子直接激活成纤维细胞,或促进转化生长因子-β表达间接发挥成纤维细胞的激活作用。本文对心肌梗死后心肌中巨噬细胞来源及其在成纤维细胞激活中的作用作一综述。
Macrophages play an important regulatory role in the inflammation and fibrosis process after myocardial infarction.Macrophages are classified into CCR2^(+)macrophages and CCR2^(-)macrophages based on the presence or absence of CC chemokine receptor 2(CCR2)expression.CCR2^(-)macrophages are classified into pro-inflammatory Ly6C^(hi)CCR2^(hi)CX3CR1^(lo)macrophages and anti-inflammatory Ly6C^(lo)CCR2^(lo)CX3CR1^(hi)macrophages according to the expression level of Ly6C.Ly6C^(hi)macrophages produce inflammatory cytokines in the early stage and acute inflammatory phase after myocardial infarction,whereas Ly6C^(lo)macrophages are recruited in the later stage after infarction,secreting anti-inflammatory cytokines and growth factors.Macrophages directly activate fibroblasts by paracrine inflammatory factors such as interleukin(IL)-1β,IL-6 and tumor necrosis factor-α,or indirectly play a role in activation of fibroblasts by promoting the expression of transforming growth factor-β.This article provides a review of macrophage sources in the myocardium and their role in fibroblast activation after myocardial infarction.
