摘要
目的研究三七粉热处理前后5种皂苷含量、水分和微生物数量变化,并建立三七粉高效液相(HPLC)指纹图谱,为三七粉热处理工艺优化及质量控制提供参考。方法设置90、95、100℃共3个热处理温度梯度,并分别在热处理时长为8、10、12 h时取样。采用HPLC法,以三七皂苷R_(1)、人参皂苷Rg_(1)、人参皂苷Re、人参皂苷苷Rb_(1)、人参皂苷Rd为指标,运用SPSS Statistics 26和SIMCA 14.1进行主成分分析(PCA)和聚类分析(HCA),同时测定各样品水分含量和微生物数量,观察三七粉在热处理前后的变化规律。结果三七粉经过不同温度和时间热处理后,5种皂苷总含量、水分含量、微生物数量在100℃热处理12 h时达到最佳。结论建立的三七粉指纹图谱测定方法准确灵敏,化学计量法能够较好地辨识影响三七粉质量的标志性成分,通过对样品5种皂苷含量、水分、微生物数量的多维度分析,整体性评价了三七粉的不同热处理工艺参数,为建立三七粉中间品内控标准提供依据。
Objective To study the changes in the content of five saponins,moisture and microbial quantity before and after heat treatment of Panax notoginseng powder,and to establish a high performance liquid phase(HPLC)fingerprint of Panax notogin-seng powder to provide reference for the optimization of heat treatment process and quality control of Panax notoginseng powder.Methods Three heat treatment temperature gradients of 90,95,and 100℃were set,and samples were taken when the heat treatment time was 8,10,and 12 h,respectively.Using HPLC method,using natoginsenostde R_(1),ginsenoside Rg_(1),ginsenoside Re,ginsenoside Rb_(1),ginsenoside Rd as indicators,principal component analysis(PCA)and cluster analysis(HCA)were per-formed using SPSS Statistics 26 and SIMCA 14.1,the moisture content and the number of microorganisms in each sample were measured at the same time,and the change rule of Panax notoginseng powder before and after heat treatment was observed.Re-sults After heat treatment of Panax notoginseng powder at different temperatures and times,the total content of the five saponins,the moisture content and the number of microorganisms reached the best when heat treatment at 100℃for 12 h.Conclusion The establishment of the fingerprint method of Panax notoginseng powder is accurate and sensitive,and the stoichiometric method can better identify the landmark components that affect the quality of Panax notoginseng powder.The different heat treatment process parameters of Panax notoginseng powder were evaluated holistically,which provided the basis for establishing the internal control standard of Panax notoginseng powder intermediate products.
