摘要
目的:探讨宁心安神中药酸枣仁-柏子仁对小鼠下丘脑5-HT能神经通路表达的影响。方法:首先,实验通过检测酸枣仁-柏子仁有效组分对小鼠海马区、下丘脑部位以及前额叶皮质区色氨酸羟化酶(TPH)和5-HT含量变化,考察酸枣仁-柏子仁有效组分对小鼠海马区、下丘脑部位以及前额叶皮质区TPH和5-HT生成的影响;其次通过TRIZOL法提取小鼠下丘脑RNA进行逆转录,最后运用Real-time PCR技术检测5-HT转运体的编码基因Slc6a4基因及5-HT1A受体编码基因Htr1a表达比值,以此探讨酸枣仁-柏子仁有效组分对小鼠下丘脑5-HT转运体编码基因及5-HT1A受体编码基因表达的影响。结果:实验结果显示,与空白组相比,给药组小鼠下丘脑部位、海马区以及前额叶皮质区TPH和5-HT的含量均显著升高(P<0.05);酸枣仁-柏子仁有效组分能使小鼠下丘脑部位Slc6a4基因表达明显降低,而Htr1a基因表达明显增加。结论:实验结果表明酸枣仁-柏子仁有效组分能通过影响色氨酸羟化酶(TPH)、5-HT转运体编码基因Slc6a4基因及5-HT1A受体编码基因Htr1a基因来增强5-HT能神经系统的效应,进而来发挥改善睡眠的作用。
Objective:To investigate the effect of effective components of Sour jujube kernels-cypress kernels Powder on the expression of 5-HT nerve pathway in hypothalamus of mice.Methods:The content of tryptophan hydroxylase(TPH)and 5-HT in different brain regions of mice was detected,and the effect of effective components of Sour jujube kernels-cypress kernels Powder on the production of 5-HT in different brain regions of mice was investigated.After retroviral transcription of mouse hypothalamic RNA extracted by TRIZOL method,the expression ratio of Slc6a4 gene encoding 5-HT transporter and Htr1a gene encoding 5-HT1A receptor was detected by Real-time PCR.The effect of active ingredients of Powder on the expression of 5-HT transporter coding gene and 5-HT1A receptor coding gene in hypothalamus of mice.Result:Compared with the blank group,the TPH and 5-HT concentration in hippocampus,hypothalamus and prefrontal cortex of mice in our jujube kernels-cypress kernels Powder group was significantly increased(P<0.05);the Slc6a4 gene expression in hypothalamus of mice was significantly decreased,while the Htr1a gene expression was significantly increased in our jujube kernels-cypress kernels Powder group.Conclusion:The experimental results show that the effective components of our jujube kernels-cypress kernels Powder can enhance the effect of 5-HT nervous system by affecting tryptophan hydroxylase(TPH),5-HT transporter coding gene Slc6a4 and 5-HT1A receptor coding gene Htr1a,thereby improving sleep.
