草原革蜱中马泰勒虫的分子检测和进化分析

Molecular detection and evolutionary analysis of Theileria equi in Dermacentor nuttalli(Acari:Ixodidae)

摘要 为厘清马泰勒虫的生物媒介及分子进化特征,对收集的新疆部分区域蜱进行基因组DNA提取,利用989/990引物对上述基因组进行梨形虫18S rRNA的PCR检测,并对阳性产物测序。利用MEGA 11.0软件包中的Alignment Explorer CLUSTAL X进行多序列比对,Bootstrap Test of Phylogeny模块下的NJ法构建马泰勒虫不同地理株的进化树。PCR检测结果显示,在收集的80只草原革蜱中有4只感染了马泰勒虫,感染率为5.00%。序列分析表明,马泰勒虫分为2个主要基因型,1个亚型。世界范围内流行的主要是Ⅰ型,Ⅱ型在南非以及苏丹等地流行。此次检测到的新疆马泰勒虫与国内四川株以及哈萨克斯坦、西班牙、沙特阿拉伯、爱尔兰分离株处于相同的进化分支,其基因型均隶属于Ⅰ型。同样的隶属于Ⅰ型的还存在1个亚型(Ⅰ-A),该基因亚型的虫株在肯尼亚和意大利均有分布。马泰勒虫18S rRNA基因V4变异区多序列比对表明,15位点Ⅱ型较Ⅰ型有A的缺失,而且在18~22位点处有TCGAG到ACAGT的突变,48位点处存在G到T的突变。上述研究表明,首次检测到草原革蜱中马泰勒虫的感染。而此次检测到的马泰勒虫隶属于马泰勒虫Ⅰ型,该基因型的马泰勒虫是世界主要流行虫株。 To clarify the ticks-vector of Theileria equi,genomic DNA was extracted from ticks,which were collected from some regions of Xinjiang Uygur Autonomous Region(XUAR),and the 18S rRNA of piroplasma were detected with primer 989/990 by PCR,and the positive products were sequenced.The CLUSTAL X software was used for multi-sequence alignment,and the Neighbor-Joining(NJ)method was used to construct the phylogenetic tree for different geographical strains of T.equi.The PCR results showed that the infection rate was 5.00%for T.equi in Dermacentor nuttalli(n=80).Sequencing analysis showed that T.equi was divided into two main genotypes and one subtype.And the Type-Ⅰwas main epidemic in worldwide,and Type-Ⅱwas mainly popular in South Africa and Sudan.The detected T.equi in XUAR was in the same evolutionary branch as Sichuan strain in China,Kazakhstan,Spain,Saudi Arabia and Ireland,and its genotypes all belong to Type-I.There was also a subtype belonging to Type-Ⅰgenotype,which was mainly prevalent in Kenya and Italy.Multiple sequence alignment of V4 variant region of 18S rRNA gene of T.equi showed that typeⅡhad a deletion of“A”at site 15,and a mutation from“TCGAG”to“ACAGT”at site 18-22 and a mutation from“G”to“T”at site 48.The above studies showed that the infection of T.equi was detected for the first time in D.nuttalli.The T.equi belongs to the Type-Ⅰin the XUAR,which is the main epidemic strain in the world.
