摘要
对SIEC贴壁细胞进行无血清全悬浮驯化。驯化的过程:选取SIEC贴壁细胞依次进行降血清贴壁培养、悬浮培养基筛选、传代驯化。驯化后的SIEC-S细胞分散好,大小均一,在CD PK15259无血清悬浮培养基(建顺生物)中稳定增殖,活细胞密度可达到1.12×10^(7)cells/mL。SIEC-S对猪流行性腹泻病毒(PEDV)敏感性高,接种PEDV后,病毒效价可达到10^(7.75)TCID_(50)/m L,适合PEDV病毒进行大规模生产。
SIEC adherent cells were acclimated by total suspension without serum.The process of domestication was as follows:SIEC adherent cells were selected for serum-reducing adherent culture,suspension medium screening,and subgeneration domestication.After acclimation,SIEC-S cells were well dispersed and uniform in size,and increased steadily in CD PK15259 serum-free suspension medium(JSBio),and the living cell density could reach 1.12×10^(7)cells/mL.SIEC-S is highly sensitive to porcine epidemic diarrhea virus(PEDV),and the titer of the virus can reach 10^(7.75)TCID_(50)/m L,which is suitable for large-scale production of PEDV virus.
