摘要
硫辛酸是一种类似于维他命的辅酶,存在于细胞中的线粒体中,可以减少并消除引起衰老和致病的自由基。因此,硫辛酸以其独特的抗氧化能力,常常作为研发抗衰老药物的重要原料药。以6,8-二氯辛酸为原料与硫脲反应生成6,8-二巯基辛酸,再与氧化剂磺化环合反应生成硫辛酸。通过探究催化剂种类、用量、反应温度与反应时间对6,8-二巯基辛酸磺化环合的影响,发现当铁盐石墨烯新型材料作为催化剂、m(6,8-二巯基辛酸)∶m(铁盐石墨烯)=1∶0.1、反应温度为60℃、反应时间为6 h时,此条件下硫辛酸的收率为97.2%,纯度为98.5%。
Alpha lipoic acid is a vitamin-like coenzyme found in mitochondria in cells that can reduce and eliminate free radicals that cause aging and disease.Therefore,with its unique antioxidant capacity,lipoic acid is often used as an important API for the development of anti-aging drugs.In this paper,6,8-dichlorooctanoic acid was used as raw material to react with thiourea to generate 6,8-dimercaptocaprylic acid,and then cyclization with oxidant sulfonated to generate lipoic acid.By exploring the effects of catalyst type,dosage,reaction temperature and reaction time on sulfonated cyclization of 6,8-dimercaptooctanoic acid,it was found that when the new iron salt graphene material was used as catalyst,m(6,8-dimercaptooctanoic acid)∶m(iron salt graphene)=1∶0.1,the reaction temperature was 60℃,and the reaction time was 6 h,the yield of alpha lipoic acid under these conditions was 97.2%and the purity was 98.5%.
