摘要
目的探讨丹白颗粒对盆腔炎性疾病后遗症模型大鼠炎症因子、子宫组织Toll样受体4(toll-like receptor 4,TLR4)/髓样分化因子8(myeloid differentiation factor 88,MyD88)/核转录因子κB p65(nuclear factor kappa-B p65,NF-κB p65)信号通路相关蛋白表达水平的影响,及其治疗盆腔炎性疾病后遗症的作用机制。方法将42只SD大鼠按照随机数字表法分为6个组,分别为空白对照组、模型组、丹白颗粒低剂量组、丹白颗粒中剂量组、丹白颗粒高剂量组、康妇炎胶囊组(阳性药对照组),以苯酚胶浆注射联合机械损伤法复制大鼠盆腔炎性疾病后遗症模型,造模成功后,空白组和模型组采用10 ml/(kg·d)生理盐水灌胃;丹白颗粒低、中、高剂量组分别用丹白颗粒药物混悬液1.25、2.5、5.0 g/(kg·d)灌胃;阳性药对照组用康妇炎胶囊混悬液0.25 g/(kg·d)灌胃。干预14 d后进行取材,通过苏木精-伊红染色法(hematoxylin-eosin staining,HE)对各组大鼠进行子宫组织学观察,酶联免疫吸附测定法(enzyme-linked immunosorbent assay,ELISA)检测大鼠外周血白介素-6(interleukin 6,IL-6)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白介素-4(interleukin 4,IL-4)、白介素-10(interleukin 10,IL10)等炎症因子水平,蛋白印迹试验(western-blot,WB)法检测TLR4/MyD88/NF-κB p65信号通路相关蛋白表达。结果HE染色结果显示,与空白组比较,模型组子宫内膜腔壁结构改变,子宫内膜充血水肿,有大量或层状性炎性细胞浸润,纤维组织增生,上皮细胞变性坏死,腺体萎缩变形;各治疗组在宫腔粘连、子宫内膜充血水肿、炎细胞浸润等方面均较模型组有不同程度的缓解,其中以丹白颗粒高、中剂量组较为显著(P<0.05)。ELISA结果显示,与空白组比较,模型组大鼠血清IL-6、TNF-α表达显著升高(P<0.05),IL-4、IL-10表达显著降低(P<0.05);与模型组比较,各治疗组IL-6、TNF-α表达均显著降低(P<0.05),IL-4、IL-10表达均显著
Objective To investigate the effects of Danbai granule on inflammatory factors and expression levels of Tolllike receptor 4(TLR4)/myeloid differentiation factor 88(MyD88)/nuclear factor kappa-B p65(NF-κB p65)signaling pathway related proteins in uterine tissue of sequelae of pelvic inflammatory disease model rats,and to explore its treatment mechanism.Methods Forty-two SD rats were divided into six groups according to the randomized numerical table method,namely,blank control group,model group,Danbai low-dose group,Danbai medium-dose group,Danbai high-dose group,and Kangwuyan capsule group(the positive drug control group),and the rat pelvic inflammatory disease sequelae model was replicated by phenol plasma injection combined with the method of mechanical injury.After the modeling was successful,the blank group and the model group were gavaged with 10 ml/(kg·d)of physiological saline;the Danbai low,medium,and high dose group were gavaged with Danbai granule drug suspension 1.25,2.5,5.0 g/(kg·d)by gavage;Kang Gynecitis capsule group was gavaged with Kang Gynecitis capsule suspension 0.25 g/(kg·d),and samples were taken 14 days after the intervention,and the uterus was observed histologically by hematoxylin-eosin staining(HE),The enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of inflammatory factors such as interleukin 6(IL-6),tumor necrosis factor-α(TNF-α),interleukin 4(IL-4),interleukin 10(IL-10)in the peripheral blood of rats,and the Western-blot(WB)method was used to detect the expression of proteins related to the TLR4/MyD88/NFκB p65 signaling pathway.Results HE staining results showed that compared with the blank group,the model group had changes in endometrial luminal wall structure,endometrial congestion and edema,massive or laminar inflammatory cell infiltration,fibrous tissue hyperplasia,epithelial cell degeneration and necrosis,and glandular atrophy and deformity;the treatment groups had different degrees of relief in terms of uterine adhesion,congestion and edema,and inflam
