摘要
氮气气调是一种安全且环保的绿色储粮技术,但害虫在长期的低氧胁迫下产生了耐受性。为明确氮气气调对储藏物害虫的防治效果和储藏物害虫低氧耐受性产生的机制,采用体积分数为95%、97%和98%的氮气分别处理赤拟谷盗Tribolium castaneum的幼虫、蛹和成虫,处理时间分别为1、3、5、7 d,记录赤拟谷盗的死亡情况和体内起保护作用的抗氧化酶的活性。结果表明,随着氮气体积分数的增大和低氧处理时间的延长,赤拟谷盗的幼虫、蛹和成虫的死亡率均逐步升高。体积分数为98%的氮气处理不同生命阶段的赤拟谷盗的死亡率显著高于95%和97%体积分数氮气下的死亡率,体积分数为98%的氮气处理7 d时的赤拟谷盗的幼虫、蛹和成虫的死亡率分别达到了98.61%、80.19%、30.55%。低氧处理后,赤拟谷盗幼虫体内的SOD、CAT和POD酶的活性均显著升高(P<0.05),但随着低氧处理时间和氮气体积分数的变化,不同酶的活性的变化趋势不同。POD酶对低氧的响应速度最快、强度最强;CAT酶对高体积分数氮气的响应速度较快,对较低体积分数氮气的响应速度较慢;而SOD酶对低氧的响应速度较POD酶和CAT酶均要慢。研究结果明确了不同体积分数的氮气对赤拟谷盗的幼虫、蛹和成虫的防治效果以及赤拟谷盗幼虫体内不同抗氧化酶对低氧的响应模式,为氮气气调技术在储藏物害虫防治中的应用提供了参考。
Controlled atmosphere with nitrogen is a safe and environmentally friendly green grain storage technology.However,the pests have developed tolerance under long-term hypoxia.To investigate the effects of controlled atmosphere with nitrogen on the control and the mechanism of hypoxic tolerance of stored product pests,Tribolium castaneum larvae,pupae,and adults were exposed to 95%,97%,and 98%nitrogen levels for 1,3,5 d and 7 d,and subsequent mortalities and the activities of antioxidant enzymes were assessed.The results indicated that with the increase of nitrogen level and the prolongation of hypoxia exposure time,the mortalities of larvae,pupae and adults increased gradually.The mortalities of larvae,pupae and adults were 98.61%,80.19%and 30.55%,respectively,after exposure to 98%nitrogen level for 7 d.After hypoxia exposure,the activities of SOD,CAT and POD enzymes were significantly increased(P<0.05).However,with the change of hypoxia exposure time and nitrogen level,the change trend of different enzyme activities was different.POD enzyme had the fastest response speed and the strongest intensity to hypoxia.The response speed of CAT enzyme to high level of nitrogen was faster,but the response speed of CAT enzyme to low level of nitrogen was slower.The response speed of SOD to hypoxia was slower than that of POD and CAT enzymes.The results clarified the control effects of different levels of nitrogen on larvae,pupae and adults,and the response of different antioxidant enzymes in larvae,providing a reference for the application of Controlled atmosphere with nitrogen technology in the control of stored pests.
