摘要
为了解2018—2021年我国部分省份牛病毒性腹泻病毒(BVDV)的流行情况,本试验采用ELISA和荧光定量PCR方法对来自河北、河南和山东等8个省份的5270份血清和1363份组织样品分别进行BVDV抗体和核酸检测,同时利用微量血清中和试验方法检测BVDV疫苗接种场采集的1449份血清的中和抗体水平。BVDV抗体检测结果显示,在8个省份采集的5270份血清样品中,BVDV抗体总阳性率为78.39%,其中河北抗体阳性率最高(90.03%),甘肃抗体阳性率最低(70.95%);2018—2021年调查区域牛群BVDV抗体阳性率呈逐年上升趋势,年阳性率分别为75.59%、75.44%、86.24%和86.62%。BVDV核酸检测结果显示,采集的1363份组织样品中BVDV核酸总阳性率为7.19%,河北最高(20.71%),山东最低(1.17%);2018—2021年调查区域牛群BVDV核酸年阳性率分别为5.78%、7.96%、3.57%和11.76%。疫苗免疫牛群的抗体持续监测结果显示,在检测的1449份牛血清样品中,BVDV中和抗体效价不低于1∶128的比例达到90.48%;2018—2021年调查牛群BVDV中和抗体效价平均值由1∶938.2上升至1∶1579.3,牛群BVDV中和抗体水平逐渐集中分布在1∶1000~1∶2000,表明疫苗提高了牛群整体的免疫状态。本试验通过调查分析国内主要养牛省份的BVDV流行情况,更新了我国BVDV的流行病学数据,结合疫苗免疫后牛群的抗体监测数据,证实BVDV在国内的流行日趋严重,加强疫病监测和疫苗预防接种将成为我国BVDV防控的关键。
In order to understand the prevalence of bovine viral diarrhea virus(BVDV)in some provinces of China from 2018 to 2021,this study used ELISA and quantitative fluorescence PCR methods to detect BVDV antibodies and nucleic acids in 5270 serum and 1363 tissue samples from eight provinces,including Hebei,Henan,and Shandong,and detected the neutralizing antibody levels in 1449 serum samples collected from BVDV vaccine immunization farms using the micro-serum neutralization test.The results of BVDV antibody detection showed that the total positive rate of BVDV antibodies in 5270 serum samples collected from eight provinces was 78.39%,with the highest antibody positive rate in Hebei(90.03%)and the lowest in Gansu(70.95%).The positive rate of BVDV antibodies in the cattle population in the surveyed areas showed an increasing trend year by year,with annual positive rates of 75.59%,75.44%,86.24%,and 86.62%from 2018 to 2021,respectively.The results of BVDV nucleic acid detection showed that the total positive rate of BVDV nucleic acid in 1363 tissue samples was 7.19%,with the highest nucleic acid positive rate in Hebei(20.71%)and the lowest in Shandong(1.17%).The annual positive rates of BVDV nucleic acid in the cattle population in the surveyed areas from 2018 to 2021 were 5.78%,7.96%,3.57%,and 11.76%,respectively.The continuous monitoring results of neutralizing antibodies in vaccinated cattle showed that among the tested 1449 cattle serum samples,90.48%had neutralizing antibody titers of not less than 1∶128.The average neutralizing antibody titer of cattle herds surveyed from 2018 to 2021 increased from 1∶938.2 to 1∶1579.3,with neutralizing antibody levels gradually concentrated in the range of 1∶1000 to 1∶2000.This indicates that vaccine immunization has improved the overall immune status of cattle herds.This study updates the epidemiological data of BVDV in China by investigating the prevalence of BVDV in major cattle-raising provinces.Combined with the antibody monitoring data after vaccine immunization,it c
