新芒果苷对小鼠磨损颗粒诱导骨溶解动物模型中炎性骨溶解影响的研究

摘要 目的探讨新芒果苷(neomangiferin,NG)对小鼠磨损颗粒诱导的炎性骨溶解的影响。方法将45只雌性BALB/c小鼠随机分为对照组、模型组和NG治疗组,每组15只,均采用手术方法建立小鼠气囊植骨骨溶解模型。对照组注射0.3 mL无菌PBS液,模型组注射0.3 mL预先处理好的钛颗粒进行建模,NG治疗组钛颗粒处理同模型组、经腹腔注射给予NG 2.5 mg·(kg·d)^(-1);连续给药14 d后处死小鼠,对气囊内植骨组织进行取材。通过HE染色检测气囊植骨组织炎性反应、观察溶骨情况;通过TRAP染色检测气囊植骨组织破骨细胞数目;通过免疫组化实验检测气囊植骨组织中白细胞介素6(IL-6)、肿瘤坏死因子-α(TNF-α)和基质金属蛋白酶-9(MMP-9)的表达情况。结果HE染色结果显示,模型组较对照组植骨气囊内植入骨块溶骨现象明显,可见明显炎性反应和大量炎性细胞浸润;NG治疗组植骨气囊内骨块较为完整,炎性反应较轻。TRAP染色检测结果显示,模型组阳性表达情况高于对照组(P<0.05);NG治疗组阳性细胞表达情况高于对照组,但低于模型组(P<0.05)。免疫组织化学染色实验显示,模型组植骨气囊中IL-6、TNF-α和MMP-9阳性表达增多(P<0.05);NG治疗组中IL-6、TNF-α和MMP-9阳性表达较模型组减少(P<0.05)。结论NG可以有效缓解磨损颗粒诱导的骨溶解动物模型中骨组织的溶骨现象与炎性反应,抑制溶骨细胞因子和炎性介质的表达。 Objective To investigate the effect of neomangiferin(NG)on inflammatory osteolysis induced by titanium particles in mouse.Methods Forty-five female BALB/c mice were randomly divided into control group,model group and NG treatment group.The osteolysis model of air pouch in mice were established by surgical method.The mice of control group were injected with 0.3 mL sterile PBS solution,and the mice of model group were injected with 0.3 mL pre-treated titanium particles for modeling.The mice of NG treatment group was treated with titanium particles as the model group,and neomangiferin(NG)2.5 mg·(kg·d)^(-1) were given by intraperitoneal injection.After 14 days of continuous administration,the mice were killed and the bone graft inside the air pouch was sampled.HE staining was used to detect the inflammatory response of balloon bone grafting tissue and observe the situation of bone lysis.The number of osteoclasts were detected by TRAP staining.The expressions of interleukin-6(IL-6),tumor necrosis factor-α(TNF-α)and matrix metalloproteinase-9(MMP-9)in air pouch were detected by immunohistochemistry.Results HE staining showed that compared with the control group,the model group had obvious osteolytic phenomenon with obvious inflammatory reaction and a large number of inflammatory cell infiltration.In the NG treatment group,the bone mass in the balloon of bone graft was relatively intact and the inflammatory reaction was less.TRAP staining showed that the positive expression of the model group was higher than that of the control group(P<0.05).The positive cell expression of NG treatment group was higher than that of control group,but lower than that of model group(P<0.05).Immunohistochemical experiments showed that the positive expressions of IL-6,TNF-α and MMP-9 increased in the model group(P<0.05).The positive expressions of IL-6,TNF-α and MMP-9 in the NG treatment group were decreased compared with those in the model group(P<0.05).Conclusion Neomangiferin can effectively relieve the osteolytic phenomenon and inf
