摘要
为探究玉兰在乌鲁木齐地区脆弱严苛环境条件下的引种栽植效果,对引种驯化7 a的望春玉兰、玉灯玉兰、紫二乔玉兰3个品种的关键气候因素、物候特性、开花动态特征等方面进行了适应性评价;以玉灯玉兰、紫二乔玉兰为试验材料,采用MTT染色法测定了不同散粉方式下的花粉活力,同时采用不同浓度梯度的硼酸、蔗糖溶液对紫二乔玉兰进行了花粉离体培养,以期选育出耐寒性、耐旱性玉兰新品种。结果表明:干旱及低温是影响玉兰栽植成功的关键因素;3种玉兰均为早春开花,开花展叶早晚顺序为望春玉兰>玉灯玉兰>紫二乔玉兰;开花过程在乌鲁木齐地区表现为花朵异步雌雄异熟;室内培养散粉的花粉活力大于室外自然盛开时的花粉活力,花粉活力最高达63.39%;紫二乔玉兰花粉离体培养最佳的溶液体系为5%蔗糖+0.001%的硼酸溶液,萌发率达53.49%。
In order to explore the effect of introduction and cultivation of Magnolia in fragile and harsh environment of Urumqi,the adaptability evaluation of M.biondii Pampan,M.denudata Desr.‘Lamp’and M.soulangeana Soul.-Bod.‘Purpurea’introduced and domesticated for 7 years was carried out from the key climate factors,phenological observation,flowering dynamic law and other aspects.M.denudata Desr.‘Lamp’and M.soulangeana Soul.-Bod.‘Purpurea’were used as experimental materials,and the pollen viability of M.denudata Desr.‘Lamp’and M.soulangeana Soul.-Bod.‘Purpurea’was determined by MTT staining method under different dispersing methods.At the same time,different concentrations of boric acid and sucrose solution were used to carry out in vitro pollen culture of M.soulangeana Soul.-Bod.‘Purpurea’,so as to prepare for breeding cold resistant varieties of Magnolia.The results showed that drought and low temperature were the key factors affecting the success of Magnolia cultivation.All three species of Magnolia bloomed in early spring,and the order of flowering and leaf spreading was M.biondii Pampan>M.denudata Desr.‘Lamp’>M.soulangeana Soul.-Bod.‘Purpurea’.The flowering process is characterized by asynchronous dioecious.The pollen vitality of indoor dry loose pollen is higher than that of natural blooming pollen,and the highest pollen vitality is 63.39%.The best solution system for in vitro culture of M.soulangeana Soul.-Bod.‘Purpurea’pollen is 5%sucrose+0.001%boric acid solution,and the germination rate is 53.49%.
