黄芪多糖辐照预处理提取工艺及活性研究 被引量:3

摘要 为实现黄芪多糖的高效提取利用,本试验以多糖提取率和DPPH自由基清除率为指标,研究黄芪多糖的辐照预处理提取工艺,比较未辐照提取黄芪多糖(APS)和辐照预处理提取黄芪多糖(RAPS)在提取率、结构、分子量和抗氧化活性等方面的差异性。结果表明,黄芪多糖的最佳辐照预处理提取工艺为辐照剂量75 kGy、提取温度70℃、提取时间2 h。微观形貌结构显示,辐照可以降解黄芪纤维结构,促进黄芪多糖的溶出;RAPS的提取率为12.56%,总糖提取率为6.97%,较APS(提取率6.78%,总糖提取率1.42%)分别提高5.78和5.55个百分点;RAPS清除DPPH自由基的IC_(50)值为2.78 mg·mL^(-1),抗氧化活性明显优于APS(IC_(50)值为5.23 mg·mL^(-1));此外,两种多糖红外结构无明显变化;重均分子量(Mw)从34.21×10^(4)(APS)降低到16.16×10^(4)(RAPS),多分散性系数无显著变化;表明该工艺具有较高的可行性。本研究结果为黄芪多糖的高效提取与活性提升提供了新方法和理论基础。 In order to achieve the efficient extraction and utilization of Astragalus polysaccharides,the extraction ratio of polysaccharide and DPPH scavenging rate were used as indicators to study the irradiation treatment extraction process of Astragalus polysaccharide.The differences in extraction rate,structure,molecular weight and antioxidant activity between untreated Astragalus polysaccharide(APS)and irradiated Astragalus polysaccharide(RAPS)were compared.The results showed that the optimum irradiation treatment and extraction process for Astragalus polysaccharide were as follows:irradiation dose 75 kGy,extraction temperature 70℃and extraction time 2 h.The microstructure showed that irradiation could degrade the the fiber structure of astragalus and promote the dissolution of the polysaccharides.The extraction ratio of RAPS and total polysaccharides after irradiation treatment was 12.56%and 6.97%,respectively,which was 5.78 and 5.55 percentage points higher than that of APS(6.78%and 1.42%).The IC_(50) value of RAPS in scavenging DPPH was 2.78 mg·mL^(-1),and the antioxidant activity was significantly higher than APS(IC_(50) value was 5.23 mg·mL^(-1)).In addition,there was no significant change in the infrared structure of the two polysaccharides.The weight-average molecular weight(Mw)of Astragalus polysaccharides decreased from 34.21×10^(4)(APS)to 16.16×10^(4)(RAPS),and the polydispersity coefficient did not change significantly.All the above indicate that the process had high feasibility.The results would provide novel method and theoretical basis for the efficient extraction and activity enhancement of Astragalus polysaccharides.
