慢性鼻-鼻窦炎患者HIF-1α、FoxM1的表达及其与黏蛋白MUC5AC、MUC5B的相关性 被引量:3

摘要 目的 探讨慢性鼻-鼻窦炎(CRS)患者缺氧诱导因子-1α(HIF-1α)、叉头框转录因子M1(FoxM1)的表达及其与黏蛋白5AC(MUC5AC)、黏蛋白5B(MUC5B)的关系。方法 选取2016年3月—2018年5月南充市中心医院行鼻内镜手术伴鼻息肉的CRS患者(CRS wNP组)和不伴鼻息肉的CRS患者(CRS sNP组)鼻窦黏膜各40例,另选取30例正常鼻腔黏膜作为对照组,采用免疫组织化学和逆转录定量PCR(qRT-PCR)检测各组HIF-1α、FoxM1、MUC5AC、MUC5B的表达,并分析它们之间的关系。结果 免疫组织化学结果显示,MUC5AC、MUC5B、HIF-1α、FoxM1在各组黏膜上皮中阳性面积均差异明显(P<0.001)。CRS wNP组和CRS sNP组各指标阳性表达率明显高于对照组(P<0.05)。qRT-PCR结果显示,各组MUC5AC、MUC5B、HIF-1α、FoxM1 mRNA的相对表达量均有明显差异(P<0.001),CRS wNP组和CRS sNP组各指标相对表达量明显高于对照组(P<0.05)。Person分析结果显示,在伴或不伴鼻息肉的CRS组织中,HIF-1α、FoxM1与MUC5AC及MUC5B mRNA的表达均存在正相关(P<0.001)。结论 发现在伴或不伴鼻息肉的CRS中,HIF-1α、FoxM1、MUC5AC、MUC5B的表达均上调,且HIF-1α、FoxM1均与黏蛋白MUC5AC、MUC5B正相关,HIF-1α和FoxM1可能在CRS中黏液生成增多中起着重要的作用。 Objective To investigate the expression of HIF-1 alpha and Foxm 1 in patients with chronic rhinosinusitis and their relationship with mucin MUC5AC and MUC5B.Methods I The sinus mucosa of CRS patients with nasal endoscopic surgery accompanied by nasal polyps(CRS wNP group)and CRS patients without nasal polyps(CRS sNP group)were selected from Nanchong Central Hospital from March 2016 to May 2018.Another 30 cases of normal nasal mucosa were selected as control group.The expressions of HIF-1α,FoxM1,MUC5AC and MUC5B in each group were detected by immunohistochemistry and reverse transcription quantitative PCR(qRT-PCR),and the relationships among them were analyzed.Results Immunohistochemical results showed that the positive areas of MUC5AC,MUC5B,HIF-1 alpha and FoxM1 in mucosal epithelium of each group were significantly different(P<0.001).In comparison to the control group,the positive expression rates of each index in the CRS wNP and CRS sNP groups were noticeably greater,and the difference was statistically significant(P<0.05).The results of qRT-PCR showed that the relative expression of MUC5AC,MUC5B,HIF1 alpha and FoxM1 in each group were significantly different(P<0.001).The relative expression of each index in CRS wNP group and CRS sNP group was significantly higher than that in control group,and the difference was statistically significant(P<0.05).Person analysis showed that HIF-1 alpha and FoxM1 were positively correlated with the expression of MUC5AC and MUC5B in CRS tissues with or without nasal polyps(P<0.001).Conclusion In CRS with or without nasal polyps,the expressions of HIF-1a,FoxM1,MUC5AC and MUC5B were up-regulated,and the mucins MUC5AC and MUC5B showed a favorable correlation with HIF-1 and FoxM1.HIF-1a and FoxM1 may play an important role in the increase of mucus production in CRS.
