山羊源加勒多尼亚链球菌(Streptococcus caledonicus)的分离鉴定及耐药性分析

摘要 为了对云南1起山羊皮下脓肿进行细菌学诊断,本研究从皮下脓肿的脓液样品中分离到2株革兰阳性链状球菌YN220769和YN220770,对2株分离株进行形态学、生理生化和16S rDNA基因鉴定并分析其药物敏感性。细菌鉴定结果显示2分离株生化特征与加勒多尼亚链球菌(Streptococcus caledonicus)模式菌株CCUG 73951T一致;与S.caledonicus参考株之间的同源性为99.6%~100.0%,与S.caledonicus CCUG 73951T之间的同源性分别为99.7%和99.9%;与5株S.caledonicus参考株形成同一个进化分支;根据鉴定结果,将YN220769和YN220770鉴定为S.caledonicus。致病性试验显示2株分离菌对小鼠有致病性。药敏试验结果显示2株分离菌对青霉素、阿莫西林、头孢噻吩等多种抗生物敏感,对磺胺甲恶唑耐药。本研究首次从山羊皮下脓肿分离到S.caledonicus,证实山羊S.caledonicus感染的存在,对山羊S.caledonicus感染的预防和控制提供了理论依据。 To bacteriologically diagnose the pathogens in a case of subcutaneous abscess in goats,two strains of Gram positive,chain-forming and coccus-shaped bacteria,designated as YN220769 and YN220770,were respectively isolated from pus of subcutaneous abscess of the necks two goats,and their morphological,physiological and biochemical and 16S rDNA gene identification and drug resistance were analyzed.The bacterial identification results indicated that the physiological and biochemical characteristics of the two isolates were consistent with that of the Streptococcus caledonicus(S.caledonicus)type stain CCUG 73951~T,and shared sequence similarity of 99.6%-100.0%with S.caledonicus reference strains,and shared sequence similarity of 99.7%and 99.9%with S.caledonicus type strain CCUG73951~T,respectively.The two strains formed the same phylogenetic clade with five S.caledonicus reference strains.Based on these results,the two strains were identified as S.caledonicus.The pathogenicity test showed that the two strains were pathogenic to mice.The drug resistance analysis indicated that the two strains were sensitive to antibiological agents such as penicillin,amoxicillin and cefalotin,and were resistant to sulfamethoxazole.To our knowledge,this is the first isolation of S.caledonicus from goat and confirmed the presence of S.caledonicus infection in goats.These results provide a theoretical basis for the prevention and control of S.ceredonicus infection in goats.
