摘要
目的:通过网络药理学为本研究构建理论基础,并构建气虚型小鼠模型,研究麦角甾醇对小鼠直肠组织中蛋白的调控作用,进一步探究其对痔形成的作用。方法:首先通过网络药理学寻找槐耳中的有效生物活性物质,探究其在人体中的生物学效应,并通过烟熏加冰水法制备气虚型小鼠模型12只,随机分为加药组和对照组,每组6只。加药组采用麦角甾醇灌胃,对照组予同等体积的有机溶剂灌胃。比较2组小鼠的直肠组织切片及蛋白表达情况。结果:网络药理学提示槐耳中的麦角甾醇与人体内类固醇类物质的合成与代谢密切相关,而类固醇类物质因其存在着明确的抗炎作用,继而可以发挥抑制痔形成及加重的效果。实验结果显示,加药组中AR蛋白表达显著低于对照组(P<0.05)。结论:麦角甾醇可以通过抑制蛋白表达而降低气虚型小鼠直肠上皮细胞中血管生成和异常增殖,存在抑制痔形成的作用。
Objective To construct a theoretical basis for this study through network pharmacology,and to construct the mouse model of qi deficiency,to study the regulatory effect of ergosterol on proteins in the rectal tissue of mice,and to further investigate its role in the formation of hemorrhoids.Methods Firstly,the effective bioactive substances in Huaier(Trametes Robiniophia Murr)were searched through network pharmacology,and their biological effects in humans were investigated.Total 12 model mice of qi deficiency were prepared by smoking and ice water method,which were randomized into the medication group and the control group,with 6 mice in each group.The mice in the medication group were treated with ergosterol by gavage,while the mice in the control group were treated with the same volume of organic solvent by gavage.The rectal tissue sections and protein expression were compared between the two groups.Results The results of network pharmacology suggested that ergosterol in Huai'er was closely related to the synthesis and metabolism of steroids in the human body,and steroids had clear anti-inflammatory effects,thereby inhibiting the formation and exacerbation of hemorrhoids.The experimental results showed that the expression of AR protein in the medication group was significantly lower than that in the control group(P<0.05).Conclusion Ergosterol can reduce angiogenesis and abnormal proliferation of rectal epithelial cells in mice of qi deficiency by inhibiting protein expression,and restrict the formation of hemorrhoids.
