川芎嗪对顺铂所致药物性耳聋大鼠耳蜗组织中自噬相关蛋白ATG调控的作用研究

摘要 目的 该实验通过观察川芎嗪(TMP)对顺铂(DDP)所致耳聋大鼠耳蜗组织中自噬蛋白ATG3和ATG7表达的影响,综合免疫组化和Western-blot的实验结果,分析TMP预防DDP所致药物性耳聋的机制。方法 购入的100只正常听力SD大鼠于动物实验饲养1周后,随机分配为盐水组、盐水+TMP组、DDP组、DDP+TMP组和3-MA组,每组各20只,盐水组和盐水+TMP组注射0.9%氯化钠溶液,共5 d;DDP组、DDP+TMP组和3-MA组按相同计量和方法及天数注射顺铂。在实验第6天,盐水组和DDP组注射0.9%氯化钠溶液,盐水+TMP组和DDP+TMP组注射TMP,3-MA组注射3-MA抑制剂,共7 d。注射第7天结束给药1 h后,收集样本,在免疫组化和Western-blot的检测方法下观察自噬蛋白ATG3和ATG7在盐水组和DDP组中的表达情况。结果 与盐水组比较,除了3-MA组,其余各组大鼠耳蜗组织中ATG3、ATG7蛋白水平升高(P<0.01);与DDP组比较,DDP+TMP组大鼠耳蜗组织中的ATG3和ATG7蛋白表达水平升高(P<0.01),而在3-MA组大鼠耳蜗组织中的ATG3和ATG7蛋白表达水平则降低(P<0.01)。结论 TMP可调控DDP诱导药物性耳聋的大鼠耳蜗组织中自噬蛋白ATG3和ATG7升高,其作用机制可能与TMP能促进自噬的发生有关。 Objective To observe the expression of autophagy proteins ATG3 and ATG7 in cochlear tissue of rats with hearing loss caused by cisplatin(DDP)treated with tetramethylpyrazine(TMP),and to synthesize the results of immunohistochemistry and Western-blot.To analyze the mechanism of TMP in preventing drug-induced deafness caused by DDP.Methods A total of 100 SD rats with normal hearing were randomly divided into saline group,saline+TMP group,DDP group,DDP+TMP group and 3-MA group,20 rats per group.Saline group and saline+TMP group were injected with normal saline for 5 days.DDP group,saline+TMP group and 3-MA group were injected with cisplatin according to the same measurement,method and days.On the 6th day of the experiment,saline and DDP groups were injected with normal saline,saline+TMP and DDP+TMP groups were injected with TMP,and 3-MA group was injected with 3-MA inhibitor for 7 days.One hour after the end of administration on the 7th day of injection,samples were collected to observe the expression of autophagy proteins ATG3 and ATC7 in saline group and DDP group by immunohistochemistry and Western-blot.Results Compared with saline group,except 3-MA group,ATG3 protein and ATG7 protein level in cochlear tissue of other groups was increased(P<0.01).Compared with DDP group,the protein expression levels of ATG3 and ATG7 in the cochlea of rats in DDP+TMP group were increased(P<0.01),while the protein expression levels of ATC3 and ATG7 in the cochlea of rats in 3-MA group were decreased(P<0.01).Conclusion TMP can regulate the increase of autophagy proteins ATG3 and ATG7 in cochlear tissue of DDP induced drug-induced deafness rats,and its mechanism may be related to the promotion of autophagy by TMP.
