摘要
目的将鹿茸与5种中药材复配成一种鹿茸复方乳液,并采用皮肤衰老模型小鼠检验该自制乳液的抗皮肤衰老效果。方法于2020年3月至2021年5月,采用注射D-半乳糖结合紫外光照射诱导小鼠皮肤衰老模型,模型小鼠按体质量均衡随机法分为空白对照组、模型组、阳性组、鹿茸复方低剂量组(5%鹿茸复方乳液)、鹿茸复方中剂量组(10%鹿茸复方乳液)、鹿茸复方高剂量组(15%鹿茸复方乳液组)、鹿茸乳液组(10%鹿茸乳液),每组10只,以同仁堂青春复颜弹力乳液作为阳性组,对照组和模型组不予给药,连续处理42 d,测各组小鼠给药部位皮肤含水率、透明质酸、羟脯氨酸、丙二醛含量、超氧化物歧化酶(SOD)活力。结果和模型组比较,鹿茸复方低、中、高剂量组均对提高衰老皮肤含水率(50.44±13.77)%比[(67.74±11.37)%、(72.76±8.13)%、(76.00±11.13)%],羟脯氨酸含量(0.16±0.21)μg/mg比[(0.35±0.12)μg/mg,(0.46±0.18)μg/mg,(0.57±0.23)μg/mg]、SOD活力(48.11±1.83)μ/mg比[(75.94±4.05)μ/mg,(107.34±4.58)μ/mg,(143.33±8.50μ/mg)]、降低丙二醛含量(9.76±6.13)nmol/mL比[(5.05±2.59)nmol/mL,(4.04±1.17)nmol/mL,(3.89±2.33)nmol/mL]有显著效果(P<0.05),中、高剂量组对提高透明质酸含量(6.87±2.34)μg/mg比[(8.44±2.20)μg/mg,(9.62±2.94)μg/mg]具有显著效果(P<0.05)。和阳性组比较,鹿茸复方低、中、高剂量组对以上指标的改善都优于阳性组。和鹿茸乳液组比较,鹿茸复方中、高剂量组对以上指标的改善相当或优于鹿茸乳液组。结论鹿茸复方乳液具有良好的抗皮肤衰老功效,10%鹿茸复方乳液抗衰老效果优于阳性药物,而与10%鹿茸乳液相当,但是更具有成本优势。
Objective In this paper,a compound emulsion was prepared with pilose antler and another five kinds of Chinese medicinal materials,and the anti-aging effect of this emulsion was tested in mice with skin aging model.Methods From March 2020 to May 2021,the skin aging model of mice was induced by D-galactose injection combined with ultraviolet light irradiation,and the mice were assigned into blank control group,model group,positive group,low-dose emulsion group(5% pilose antler compound emulsion),medium-dose emulsion group(10% pilose antler compound emulsion),high-dose emulsion group(15% pilose antler compound emulsion)and pilose antler emulsion(10% pilose antler emulsion)according to body mass based stratified random sampling method,10 mice in each group.Tongrentang youth elasticity emulsion was used for the positive group.The control group and model group were not given medication.All these treatments lasted for 42 days.Then the skin moisture,hyaluronic acid,hydroxyproline,malondialdehyde content and superoxide dismutase(SOD)activity of each group were measured.Results Compared with model group,low-,medium-and highdose pilose antler compound emulsion groups had significant effect on increasing moisture content[(50.44±13.77)%vs.(67.74±11.37)%,(72.76±8.13)%,(76.00±11.13)%],hydroxyproline content[(0.16±0.21)μg/mg vs.(0.35±0.12)μg/mg,(0.46±0.18)μg/mg,(0.57±0.23)μg/mg],SOD activity[(48.11±1.83)μ/mg vs.(75.94±4.05)μ/mg,(107.34±4.58)μ/mg,(143.33±8.50)μ/mg]of the aging skin,and decreasing the content of malondialdehyde[(9.76±6.13)nmol/mL vs.(5.05±2.59)nmol/mL,(4.04±1.17)nmol/mL,(3.89±2.33)nmol/mL](P<0.05).Besides,the medium-and high-dose groups had significant effect on increasing the content of hyaluronic acid[(6.87±2.34)μg/mg vs.(8.44±2.20)μg/mg,(9.62±2.94)μg/mg](P<0.05).Compared with the positive group,the improvement of the above indexes in the low-,medium-and high-dose groups of pilose antler compound emulsion was better.Compared with the pilose ant ler emulsion group,the improvement of
